Objective: To investigate the expression of adenkylate kinases 2 (AK2) in the brain tissues of the pharmacoresistance epilepsy patients, and to explore the possible role in pharmacoresistance epilepsy.Methods: Gene-chip,immunohistochemistry and immunofluorescence were used to test expression of AK2 mRNA and protein levels in the surgically removed brain tissue of patients with drug-refractory epilepsy from the brain bank of our department(n=40), and the results were compared with that of normal controls (n=20).Results:1.Gene chip: There was significant difference between the control group and the pharmacoresistance group (Ratio=3.209). The expression of AK2 gene was shown to be increased in the brain tissues of the pharmacoresistance epilepsy group.2.Immunohistochemical analysis: AK2 protein expression was significantly increased in the brain tissues of patients with drug-refractory epilepsy as compared with controls in the same regions.3. Immunofluorescence analysis: In the temporal lobe cortex and hippocampus, strong immunoreactivity for the staining of AK2 protein was observed. There were more obvious in the pharmacoresistance epilepsy group than in control group.Conclusions:The results indicate that the overexpression of AK2 may be involved in the drug-refractory epilepsy.
