Realgar belongs to mineral drugs with yellow arsenic,the main element As₂S₂,which has effect such as clearing away toxic and killing parastes, eliminating dampness and removing phlem,etc.It can cure some stubborn diseases,especially be applied to informal prescription or new therapy. Realgar always acts ingredient in many Chinese patent medicines,but it has arsenic which is harmful for human body.Due to the uncertain dosage and excess time,Chinese patent medicines may induce to intoxication.So it is necessary to establish lasting,exact,sensitive and convenient test on Realgar in biological specimen,study on pharmacokinetics and monitors on therapeutic drug by determining content of arsenic in body floid,the results can offer theoretic base for reasonable application and monitoring.The thesis researches pharmacokinetics character and disposision in rat's organism and in saliva of Realgar with high(7.00g/kg),low(3.50g/kg)dosage; determins concentration of arsenic in whole blood and organism by double channel atomi fluorescence spectrometry with hydride generation techinque; sets up the method of arsenic content in saliva by graphite furnace atomic fluorescence spectrometry first time;draws the clasma concentration-time curve,fits compartment model and calculates pharmacokinetics parameter.Technology investigation indicates that the methods of double channel ato -mi fluorescence spectrometry with hydride generation technique and graphite furnace atomic fluorescence spectrometry have merit such as high sensitivitymin detection quantity0.005μg/mL,good precisionRSD＜15%,good linear rela -tion between 5μg/L～80μg/L.Pharmacokinetic character of arsenic coincides with open extravenous administration sing -al compartment model,first order processes,linear kinetic character.The more administration dosage,the more concentration of arsenic in organis -um.The character of arsenic quick distribu -tion in organisum,slow clearance,will lead to it be accumulated.The distribu -tion of arsenic in saliv -a is concerned with dosage.The concentration of arsenie has dependablity between in whole blood and saliva(P＜0.05),so realgar is monitored in saliva instead of in whole blood.
