| Objective: This thesis is aimed from the standpoint of chemical monoids to study the changing pattern of acidic and alkaline components of rhubarb and Cortex Phellodendri, "the acid-alkaline pair drugs" in the Rhubarb-Saltpeter Decoction, to determine the sovereign the minister ingredient compatibility combination relation of rhubarb and Cortex Phellodendri in the recipe and the best extract method of the Decoction; to make clear the macromolecule complex salt solubility in simulated gastric fluid and simulated intestinal fluid produced by "the acid-alkaline pair drugs" of rhubarb and Cortex Phellodendri. The differences of pharmacodynamic effects with changes of acidic and alkaline components were inspected and verified by animal experiments. The research is expected to direct clinical application of "the acid-alkaline pair drugs" of rhubarb and Cortex Phellodendri, and will be beneficial to the research of TCM compatibility from chemistry concern.Method: The anthraquinones and the berberine hydrochloride in samples of different extract methods and different compatibility combinations of "the acid-alkaline pair drugs" rhubarb and Cortex Phellodendri were determined by HPLC. The precipitates of the water decoctions of "the acid-alkaline pair drugs" of rhubarb and Cortex Phellodendri were dissolved in simulated gastric fluid and simulated intestinal fluid, and main chemical components were determined. The TCM jaundice of Yang-Huang animal liver damage model was induced byα-ANIT with high-fat-glucose diet in a damp and warm environment. The rats were administered by oral with the Rhubarb and Saltpeter Decoctions of different extract methods, the general conditions of the rats were observed, TBIL,ALT,AST in serum were detected, and liver slide were examined.Result: Among the four kinds of different extract methods of the Rhubarb-Saltpeter Decoction, the anthraquinones were higher in water extracts than in that of in ethanol extracts. It concluded that water extract method is superior to that of ethanol extract in different compatibility assembles, the content of the anthraquinones in water decoction was the maximum.The berberine hydrochloride of single herb in water extracts is more than that of ethanol extracts in samples of four different extract methods of the Rhubarb-Saltpeter Decoction; the highest amount was detected in water decoction. In a similar way, the berberine hydrochloride in pair drugs and Rhubarb-Saltpeter Decoction of ethanol extracts is more than that in water extracts, and highest in ethanol reflux liquid.In different compatibility combinations, the contents of anthraquinones and berberine hydrochloride in samples of water extracting solutions of the single herb were far more than that's in pair drugs and Rhubarb-Saltpeter Decoctions, and there were no significant difference between Pair drugs and Rhubarb-Saltpeter Decoction. Second, it was showed no significant difference in the contents of the anthraquinones and the Berberine hydrochloride in ethanol extracts among single herb, pair drugs and whole recipe. Among all the compatibility combinations and extract methods, the highest contents of the anthraquinones and the berberine hydrochloride were detected in water extracting solution of single herbs.Between water extract solution and ethanol extract solution, the contents of the anthraquinones and the berberine hydrochloride with proportion of the Rhubarb-Saltpeter Decoction were very nearly in the water extract solution, and the content of the anthraquinones was far lower than that of the berberine hydrochloride in the ethanol extract solution. Simultaneously, pharmacodynamic test inspects and verifies that the effective of water extract solution is superior to that of the ethanol extract solution.The precipitation of "the acid-alkaline pair drugs" rhubarb and Cortex Phellodendri water decocting can be dissociated and dissolved by the simulated gastric fluid and the simulated intestinal fluid, and detected the contents of the anthraquinones and the berberine hydrochloride. After calculation, the contents of the anthraquinones and the berberine hydrochloride were dissolved from the simulated gastric fluid and the simulated intestinal fluid which was far lower than that of the difference value between contents of decoction and centrifugat, it proves that macromolecule complex salt in the precipitate was only dissolved a small section in the simulated gastric fluid and the simulated intestinal fluid, but a majority of macromolecule complex salt can't be dissolved.The different extract methods of the Rhubarb-Saltpeter Decoction has the varying degree therapeutic action to the TCM jaundice of Yang-Huang animal liver damage model, the performance is the anus temperature were dropped, the weight were gained, the seroenzyme ALT,AST and serum total bilirubin TBIL were reduced and the hepatic cells impairment were alleviated. It was showed the water decocting healing efficacy is optimum, but the ethanol reflux liquid is worst by comparing comprehensively different extract methods.Conclusion: The acidity and alkaline contents changing law of "the acid-alkaline pair drugs" rhubarb and Cortex Phellodendri in the Rhubarb-Saltpeter Decoction is: effective ingredient extraction quantity was detracted from "the acid-alkaline pair drugs" rhubarb and Cortex Phellodendri decoction, and should produce macromolecule complex salt precipitate which was insoluble in water. The structure of macromolecule complex salt which Rhein and Jamaicin was together decocted to generate was firstly identified by quantum chemistry method. It proves that the macromolecule complex salt formation had certain direct relation with acidity and alkaline contents interaction of "the acid-alkaline pair drugs" rhubarb and Cortex Phellodendri herbal decoction and also proves that macromolecule complex salt solubility in the simulated gastric fluid and the simulated intestinal fluid was very low; biological effective availability of macromolecule complex salt was very low by pharmacology experimental verification. When "The acid-alkaline pair drugs" rhubarb and Cortex Phellodendri used ethanol extracting method, the extraction quantity was detraction, this loss had no certain relations with each correlative herbal medicine, but had a relation with menstruum extraction.The sovereign ingredient and the minister ingredient compatibility combination relation of Rhubarb and Cortex Phellodendri in the Rhubarb-Saltpeter Decoction: it was showed from the pharmacology experiments that the Rhubarb-Saltpeter Decoction can treat the TCM jaundice of Yang-Huang animal model, thereby verify the rationality of the Rhubarb-Saltpeter Decoction compatibility combination. We adopt the pharmacology experiments to study the acidity and alkaline contents of "the acid-alkaline pair drugs" rhubarb and Cortex Phellodendri in the proportion of whole recipe, we raise an issue that rhubarb is sovereign ingredient and Cortex Phellodendri is minister ingredient for the first time.Optimum extraction methods of the Rhubarb-Saltpeter Decoction: research on the acidity and alkaline contents changing of "the acid-alkaline pair drugs" rhubarb and Cortex Phellodendri was showed that "the acid-alkaline pair drugs" rhubarb and Cortex Phellodendri decoction can lead to acidity and alkaline contents decrease and pharmacodynamic effect weakness, so it can be arrived at a conclusion that decoction with rhubarb and Cortex Phellodendri method is not perfect one. For the purpose of increasing ingredient healing effect and saving drugs, we should improve traditional decoction method. We believe that rhubarb and Cortex Phellodendri to be decocted alone was an optimal method in "the acid-alkaline pair drugs" rhubarb and Cortex Phellodendri prescriptions, but we should subject to further in-depth study on this item. |
