ObjectiveTo investigate the expression of Aurora-B in osteosarcoma,and provides a theoretical basis for finding a new molecular therapeutic target for osteosarcoma.MethodsThe expression of Aurora-B was detected by immuno-histochemistry method in tumor tissues of 40 patients with osteosarcoma. The results was compared to that in osteochondroma.ResultsThe positive stain of Aurora-B in the nucleus.Aurora-B expression was observed in osteosarcoma and osteochondroma, but the positive expression rate of Aurora-B in osteosarcoma tissues was 70% which was significantly higher than that in osteochondroma tissues (25%),(P=0.001<0.05).The expression of Aurora-B in osteosarcoma was not associated with sex(P>0.05).ConclusionsThe Aurora kinase family is a collection of highly related serine/threonine kinases that are key regulators of mitosis. These kinases are involved in multiple facets of mitosis, including centrosome duplication, mitotic spindle formation, chromosome alignment upon the spindle, mitotic checkpoint activation, and cytokinesis. Errors in these processes ultimately lead to tumor or cell death. Aurora-B is located on chromosome 17p13.1, a region not typically amplified in human malignancies. Despite lack of amplification at the gene level, mRNA and protein levels of Aurora-B are frequently increased in tumors, such as colorectal cancer. Aurora kinase is a potential molecular therapeutic target for tumor, selective inhibitor of Aurora B kinase can inhibits human tumor growth by inducing apoptosis.The expression of Aurora-B kinase in osteosarcoma has not been reported since now. The overexpression of Aurora-B in osteosarcoma possibility indicate that Aurora-B may be played an important role in the genesis and development of osteosarcoma and that Aurora-B can be a new therapeutic target for osteosarcoma.
