1. Objective:To investigate the relationship between the BSEP(bile salt export pump) gene polymorphism of pregnant women and patients with ICP(intrahepatic cholestasis of pregnancy) in jiangxi Han population .2. Methods:The samples of this study was collectd from 60 women with intrahepatic cholestasis of pregnancy and 60 normal pregnant women,two single nucleotide polymorphism(SNP) sites rs473351 and rs2287622 in BSEP gene were analysed by polymerase chain reaction-restrcition fragment length polymorphism method.The frequency of genotype and allele was performed Hardy-Weinberg equilibrium test and was compared between ICP and control group.Statistical analysis was conducted with Pearson chi-square test(SPSS13.0 software) or Fisher exact probability,the level of statistical significance was defined as P<0.05.3. Results:3.1 The genotype of SNP site rs473351 in BSEP gene all were GG genotype, we found that there were no polymorphism in all the samples.3.2 The frequency of CC,CT,TT genotype( SNP site rs2287622) was 78.3%,18.3%,3.3%in the ICP group,while was 56.7%,33.3%,10.0% in control group, We found that the constituent ratio of GT,GT,TT genotype between the ICP group and control group had significant difference(Fisher exact probability was 0.033 p<0.05) , at the same time , the C allele frequency in the ICP group were 87.5%,higher than in the control group(73.3%) (P=0.004<0.05), the two groups had statistical significance too. 4. Conclusion:We did not find rs473351 polymorphism in the BSEP gene in the jiangxi Han population,and there may not exsist relationship between ICP and rs473351 site,while the SNP site rs2287622 polymorphism(C/T) may be associated with ICP in jiangxi Han population,Women with CC and CT genotype have the higher hereditary susceptibility to ICP . C alleles may result in ICP through interfering acid bile eccrisis in liver.
