| Objective: To observe the expression of nm23 and c-myc protein in colorectal carcinoma and evaluate their clinical significance and the correlation between the expression of nm23 and that of c-myc protein in colorectal carcinoma .Methods:Forty cases of colorectal carcinoma and fifteen cases of normol tissues of the Sencond Affiliated Hospital of Nanchang University from October. 2006 to April .2007 were obtained . These tumors were made into paraffin blocks, and then to pathological sections.All patients were not treated with radiotherapy or chemotherapy. The expressions of nm23 and c-myc were detected by immunohistochemical method (Elivision Method) . We had negative control by repeating the same procedure with PBS balanced solution replacing No.1 antibody, and had positive control by repeating the same procedure with positive slice containing objective antigen. We determined sexuality, age, happening place, anteroposterior diameter, lympth node metastasis, liver metastasis, differentiation degrees and staging according clinical data. They were regarded as grouping variables, and ranking expressions of protein were regarded as index ones. All data were analysed with statistic method.Results:1.In colorectal cancer the positive expression of nm23 and c-myc protein among men respectively were 65.38%, 84.62% and those among women were 64.29%, 78.57%; those among people younger than fifty were 66.67%, 88.89% and those among people older than fifty were 63.64%, 77.27%; those in colon carcinoma were 65.38%, 80.77% and those in rectal cancer were 64.29%, 85.71%; those in tumor with diameter larger than 5cm were 66.67%, 79.17% and those in tumor with diametersmaller than 5cm were 62.50% , 87.50% ; those with lympth node metastasis were 41.18%, 94.12% and those without lympth node metastasis were 82.61%, 73.91%; those with liver metastasis were 40.00%, 93.33% and without liver metastasis were 80%, 76%; those in tumor well-differentiated were 92.86%, 64.29% and those in tumor poorly differentiated were 41.18%, 94.12%; those in A/B stage were 86.36%, 77.27% and those in C/D stage were 38.89%, 88.89%. The positive expression of nm23 and c-myc in colorectal cancer tissue were 65%, 82.50% and those in normal tissue were 93.30%, 33.33%.2.The expression of nm23 and c-myc protein were not correlated with the patient sex, age, happening place and size of the tumor respectively (P﹥0.05). On the contrary, they were significantly associated with lympth node metastasis, liver metastssis, differentiation degrees and the Dukes stage(P﹤0.05).3.The correlation between nm23 and c-myc protein expression was checked with Spearman method and had negative relationship(r=-0.322,P=0.043<0.05). Conclusion:1.The positive expression of nm23 or c-myc is obviously associated with lympth node metastasis, liver metastasis, differentiation degrees and the Dukes stage.2. The expression of nm23 and c-myc hve negative relationship.3.We may offer some bases at a molecular level for the prognosis in patients with colorectal carcinoma by detecting jointly the expression of nm23 and c-myc protein. We may offer theoretical bases to some extent for gene therapy to colorectal carcinoma. |
