Liver is the maximal parenchymatous organ in abdomen,which has important physiologic function. Hepatic injury is a common clinical symptom. Without pertinent treatment,it will induce kinds of neopathy,such as,infection, bile leakage on liver wound surface, secondary hemorrhage. It can even result in hepatic fibrosis,hepatic cirrhosis and tumor of liver,harms human health severely.There is no clinical reliable medicine to prevention and cure liver injury recently.Gynostemma pentaphyllum (Thunb.) Makino, a member of Cucurbitaceae family, is a kind of herb. The modern pharmacology research shows that Gynostemma pentaphyllum has a kind of pharmaco-activity, such as, antitumor, reducing blood lipids, hypoglycemic action, and antisenility. The present researchs are focused on Gynostemma pentaphyllum Saponins. The reseach about the other component is very few.On the fundament of preliminary experiment, we separated and purified the raw Gynostemma Pentaphyllum Polysaccharides, analysised the structure and characters. Taking HL-7702 cell, primary cultured hepatocytes in rats and rats as the research object, carbon tetrachloride and ethyl alocohol were chosen to form the chemical injury model. We studyed the protective effect both at cell and animal level.The activities of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST), malondialdehyde (MDA), superoxide dismutase (SOD) or reduced glutathione hormone(GSH), as well as the pathological changes of the liver were determined in all groups. The main contents of the papers are as follows:1. The separation,purification and the character analysis of Gynostemma Pentaphyllum Polysaccharides.After initial purification the raw Gynostemma Pentaphyllum Polysaccharides was separated by DEAE-Sephrose column and Superder G-200 culumn respectively, then desalted by Sephadex G-25, we get the fine purified Gynostemma Pentaphyllum Polysaccharides with the name of GPS-2, GPS-3, GPS-4 by freeze drying at last. As the content of GPS-4 is very less, we did not do further reseach. we did initial reseach to GPS-2 and GPS-3 about their purity,molecular weight.2. The protective effect of Gynostemma Pentaphyllum Polysaccharides on chemical liver injury in HL-7702 cell.In this paper we studyed the protective effect on chemical liver injury of GPS-2 and GPS-3 with HL-7702 cell. It is proved that GPS-3 can protect the HL-7702 cell from chemical liver injury evidently, but the protective effect of GPS-2 against chemical liver injury is not remarkable. So in latter experiment research, we used GPS-3 only to study the protective effect against chemical liver injury.3. The protective effect of Gynostemma Pentaphyllum Polysaccharides on chemical liver injury in the primary cell of liver.As HL-7702 cell is immortalization hepatic cell strain, it has many difference compared with normal liver cell, even though it has part of hepatocellular characters. So we continued to stude the protective effect on chemical liver injury of GPS-3 with the primary cell of liver.4. The protective effect of Gynostemma Pentaphyllum Polysaccharides on chemical liver injury in mice.In this paper we continued to study the protective effect on chemical liver injury of GPS-3 on whole animal level. The biochemical indicators and pathology slice indicated that GPS-3 have conspicuous effect on chemical liver injury in mice.To sum up all the experement, we know that GPS-3 has favourable protective effect against chemical liver injury, both in cellular leverl and animal level. It is a biological active component with extensive research and development value. Further reseach of the mechanism of its protective effect are expected.
