Objective:To study the antioxidation of grape seed extract on the rat model of oxidative damage byγ-irradiation,the middle-aged and elderly people.And to investigate the anti-oxidative effect of different dose grape seed extract in human body.Methods:1.In the animal study,fifty Kunming mice were randomly divided into model group, low dose group,medium dose group,high dose group and normal control group.The model group and the normal control group were given vegetable oil,the three dose groups were given grape seed extract at dose of 0.083 mg/kg.BW/d,0.167 mg/kg.BW/d,0.500 mg/kg.BW/d.The erythrocyte SOD activity,blood GSH-PX activity were detected one month later.Thenγ-irradiation was given except the normal control group,4 days later the SOD activity,GSH-PX activity and MDA content in the mice liver were measured.2.In the human study of RNI dose,110 middle-aged and elderly people were randomly divided into control group and test group.The test group was given grape seed extracts at a dose of 1g/d,equivalency with 100mg of proanthocyanidin.The control group was not treated with anything.The changes of the serum SOD activity, GSH-PX activity and serum MDA content before the study and 3 months later were studied to observe the antioxidation of grape seed extract to human body.3.In the anti-oxidative effect of different dose grape seed extract study,28 healthy volunteers aging from 60 to 65 were distributed randomly and equally into 4 groups: control group and low dose,middle dose,high dose group,according to their SOD,GSH-PX and MDA levels in serum.Normal control group was given nothing.Each dose group was given grape seed extract capsule equivalency with 50mg,100mg or 150mg of proanthocyanidin for one month.The SOD,GSH-PX and MDA levels in serum were measured both prior to and after the experiment.Results:1.In the animal study,after given grape seed extract 30 days,the erythrocyte SOD activity significantly increased in the medium dose and high dose groups compared with the model group(P<0.05),the erythrocyte SOD activity significantly increased in the medium dose group compared with the normal control group(P<0.05).The erythrocyte SOD activity increased in the low dose group and high dose group compared with the normal control group,but there was no significant difference(P>0.05).30 days after the grape seed extract were given,the blood GSH-PX activity significantly increased in the high dose group compared with the model group and the normal control group(P<0.05).And compared with the normal control group,the low dose group were higher(P<0.05).There was no significant difference of the blood GSH-PX activity between the low,medium dose group and the model group(P>0.05).There was also no significant difference of the blood GSH-PX activity between the medium dose group and the normal control group.4 days afterγ-irradiation were given,the liver SOD activity was significantly lower in the model group compared with the normal control group(P<0.05).The liver SOD activity was higher in the three dose groups compared with the model group,but only the high dose group has sense(P<0.05).The liver GSH-PX activity was significantly lower in the model group compared with the normal control group(P<0.05).The liver GSH-PX activity was significantly higher in the high dose group compared with the model group(P<0.01).The liver GSH-PX activity was higher in the low and the medium dose group compared with the model group,but there was no significant difference(P>0.05).The liver MDA content was significantly higher in the model group compared with the normal control group(P<0.05).The liver MDA content in the three dose groups were significantly lower than the model group(P<0.01).2.During the 3 months human study,9 persons quitted for various reasons.There was no significant difference of the serum SOD activity,the serum GSH-PX activity and the serum MDA content between the test group and the control group before intervention(P>0.05).There was no significant difference of the SOD activity,the GSH-PX activity in serum and the serum MDA content in the control group before and after the intervention(P>0.05).There were significant differences in all the three indexes between the test group and control group after the intervention(P<0.05,P<0.05,P<0.01).In the test group,there was significant difference of the serum SOD activity before and after intervention in the test group(P<0.01),which increased from 64.11±12.49 U/ml to 69.93±14.75 U/ml.The serum GSH-PX activity increased from 157.22±21.92 U/ml before intervention to 165.65±18.72 U/ml after intervention, which showed significant difference(P<0.01).The serum MDA content decreased from 3.87±0.98 nmol/ml before intervention to 3.25±0.82 nmol/ml after intervention, which also showed significant difference(P<0.01).3.1 month after the different dose grape seed extract were taken,There was no significant difference of all the three indexes in the control group before and after the intervention(P>0.05).After the intervention,the serum SOD activity was significantly higher in the high dose group(P<0.05).And the GSH-PX activity in serum and the serum MDA content in the medium and high dose groups before and after the intervention showed significant difference(P<0.05,P<0.01).Conclusions:1.The animal study shows that high dose of grape seed extract have the strongest effect of anti-oxidation.It can significantly increase the erythrocyte SOD activity and the blood GSH-PX activity,it can also significantly increase the liver SOD activity and the liver GSH-PX activity after given theγ-irradiation.It can significantly decrease the high liver MDA content induced by theγ-irradiation.2.The grape seed extract shows very strong effect of anti-oxidation and had no adverse reaction in the human study.It can significantly increase the serum SOD,GSH-PX activity,and also significantly decrease the serum MDA content in the test group.3.The anti-oxidative effect of different dose grape seed extract study shows that low dose of grape seed extract(50mg/d)have no significantly effect of anti-oxidation in short term.Medium dose and high dose(100mg/d,150mg/d)have significantly effect of anti-oxidation.So it is suitable to take grape seed extract equivalency with 100mg to 150mg of proanthocyanidin every day.
