Objective:Tumor is one of the most familiar diseases which threaten life and damage health of human and it is a major cause for man to death. Because lacking the effective methods to cure this disease, many of the patients with tumor get bad results. Radiotherapy is one of the routine methods. Radiotherapy can kill the tumor cell. But it can also caused the side effects on normal tissue and organ. So it is important study in radiotherapy research to kill the tumor cell in maximum with less side effects on normal tissue.Acute esophageal injuries by irradiation is one of common complications after irradiation to thoracic malignancies and head and neck cancer. However, the former researches related to acute esophageal injuries by irradiation mainly concentrated on clinical symptom and histomorphology. But these studies with clinical symptom and histomorphology had certain subjective and limitative, so they can't make quantitative analysis and objective quantization when evaluating the effect of drug, and can't exactly compare the effect of drugs. Therefore, it is urgent to further discuss the occurrence and developing mechanism of acute esophageal injuries by irradiation from the aspects of oxidative damage and inflammatory cytokine for improving grading standards of acute esophageal injuries by irradiation and objectively evaluating the effect of drugs.Traditional chinese medicine treats esophageal inuries by irradiation with considerable curative effect, and there are not severe adverse reaction. Danzhen oil is a kind of experienced prescription developed by Professor Shi Hengjun specially for esophageal inuries by irradiation. According to the clinical practical researches for many years, Danzhen oil has been found to be able to effectively relieve the clinical symptom of esophageal injuries by irradiation.In this study, firstly, using Chinese rabbits to establish the model of acute esophageal injuries by irradiation. Then preliminary studying the occurrence and developing mechanism of acute esophageal injuries by irradiation from the aspects of free radical damage and molecular biology. And observing the protective effect of Danzhen oil to on acute esophageal radiation injuries, and studying its effective mechanism, in order to provide the previous basis for conducting system research of its characteristics and action mechanism and the theoretical basis for clinical application of Danzhen oil. The experiment was divided into 3 parts.Methods and Results:In Experiment 1, the occurrence and developing mechanism of acute esophageal injuries by irradiation was preliminarily studed from the aspects of free radical damage and molecular biology. Methods: The esophageal tissue of rabbits were irradiated with 15Gy in single dose. The pathological changes of esophagus were observed by light microscopy and the level of malondialdehyde (MDA) and the expression of IL-1βand TNF-αmRNA in esophagus tissue were measured at days 1, 4 or 7 after irradiation. Results: The mucous edema and inflammation changes in esophageal tissue started on the 1stday and continued to the 7th day after irradiation, but on the 7th day basal cells have been found to proliferation and repairation. The level of MDA in esophagus was increased by day 7 after irradiation (P<0.05). And the increased expression of IL-1βand TNF-αmRNA in esophagus tissue was also observed by day 7 after irradiation.In Experiment 2, the effect of Danzhen oil to on acute esophageal radiation injuries was obeserved and its effective mechanism was studied. Methods: Eighteen rabbits were randomly allocated to 3 groups: control group, irradiation group, Danzhen oil group. The pathological changes of esophagus in rabbits were observed by light microscopy and the level of malondialdehyde (MDA), activity of superoxide dismutase (SOD), and the expression of IL-1βand TNF-αmRNA in esophagus tissue were measured after irradiation. Results: In Danzhen oil group, though vasodilatation in submucosa can still be observed, the less inflammatory cells infiltrated and obvious thickening in mucosa than irradiation group. The level of MDA and expression of IL-1βand TNF-αmRNA in esophagus tissue in irradiation group were higher compared with normal group(P<0.05). In Danzhen oil group the level of MDA and the activity of SOD was increased in Danzhen oil group(P<0.05).Meanwhile expression of TNF-αmRNA were significantly reduced contrasted with irradiation group(P<0.01).Conclusions:Through replication model, we preliminarily deduced that both lipid peroxidation of biological membrane and the changes of the expression of inflammatory cytokines such as IL-1β,TNF-αparticipate the occurrence and developing process of acute esophageal injuries by irradiation. And Danzhen oil can relieve the damage of esophageal tissue by irradiation by improving anti-oxidative capacity, removing the active oxyradical, decreasing lipid peroxidation of biological membrane, and reducing the expression of TNF-αmRNA.
