Objective:Objective to investigate the early expression of matrix metalloproteinase-2,12 after actue spinal cord injury,applies the different MMPs inhibitor to change MMP-2,12 expression level observes whether can reduce the ASCI secondary damage,and further study whether methylprednisolone intervention after acute spinal cord injury has an ability to inhibit metalloproteinase-2,12 or not.thus through the series experiment confirmed MMP-2, 12 may consider is uses in intervening treats ASCI the target member.Methods The models of spinal cord injury in rabbits were induced with Allen'S way and the expression of matrix met-alloproteinase-2,12 in injury spinal cord was observed with elisa,the tissue sections were stained with hematoxylin and eosin(HE), immunohistochemistry and Real-Time PCR,RT-PCR.The Tarlov scales of hindlimb movement were carried out.The correlation about expression of MMP-2,12 with the degree of injury were analysed.Results The expression of matrix metalloproteinase-2 incerased 2h after spinal cord injury. Which reached its peak 2 days after spinal cord injuyr(ASCI).The matrix metalloproteinase-2 positive cells were mainly expressed on ischemia neuron, angioendothelila cells and neutrophile.The expression of matrix metalloproteinase-12 incerased 2h after spinal cord injury.Which reached its peak 24h after spinal cord injuyr(ASCI),The express of matrix metallopmteinase-2,12 wasn't found in the sham-operated rabbits.The blood serum EL1SA examination demonstrated similarly MMP-2, 12 expressions compares the control group obvious ascension,There is significantly positive correlation between the level of expression of MMP-2,12 and the dgree of spinal cord injuries.Simultaneously application large dose MP and the DOX intervention,can cause in the blood serum and organization's MMP-2,12 expressions drops obviously,the promotion nerve function restores,still might decline MMP-2,12 in the ASCI wound latter 72 hours using the DOX intervention.Conclusions After ASCI in the rabbit spinal cord organization and the blood serum MMP-2, 12 expresses the ascension,the above two kinds express excessively the factor participated in ASCI,and damages certain function to the secondary,also with the ASCI degree present correlation,MP and the DOX intervention may reduce MMP-2,12 expressions obviously, simultaneously reduces the sequential damage,therefore the animal experimentation prompts MMP-2,12 is possibly a latent treatment target spot.
