The aim of this study is to isolate antibacterial constituents from the fermentation broth of 095407 from soil of mangrove plant Sonneratia paracaseolaris and endophytic fungus Penicillium citrinum Thom 093503 from mangrove plant Acrostichum aureurm L. The separation procedure was guided by antibacterial bioassay. Twenty compounds were isolated and purified by column chromatography. (3R*,4S*)-6,8-dihydroxy-3,4,7-trimethylisocou-marin (1), a new compound and 3,4,5-trimethyl-1,2-benzenediol (2), a new natural product, were identified by physicochemical and spectral data. Other compounds were identified asβ-sitosterol (3), oleic acid (4), decarboxydihydrocitrinone (5), phenol A (6), formononetin (7), uridine (8), cyclo (Ala-Ile) (9), cyclo (Gly-Tyr) (10), (3β,4β,22β)-olean-12-ene-3,22,23-triol (11), 2,3-dihydroxypropyl oleate (12), 1-(2-Furanyl)-1,2-ethanediol (13), liquiritigenin (14), cyclo (Pro-Thr) (15), cyclo (Pro-Tyr) (16), n-butyl-O-β-D-fructopyranoside (17), D-mannitol (18), 2,3-Butan ediol (19) and 4′,7,8-trihydroxyisoflavone (20).Antibacterial activities of all the compounds and the fractions were assayed by paper disco diffusion method, and compounds 2, 4, 5, 14, 15, 16 and fractions a-2, a-3 of 093503 showed inhibitory effect on Staphylococcus aureus and methicillin-resistant Staphylococcus aureus. Fractions a-1 and a-3 of 093503 showed inhibitory effect on Candida albicans. The majority of compound from fractions a-1, a-2 and a-3 were elucidated by GC-MS. a-1 was consist of methyl palmitate, methyl oleate and methyl stearate. a-2 was made up of methyl stearate and methyl linoleate. The constituents of a-3 were oleic acid, benzoic acid and phenylacetic acid. Antibacterial activities of 2, 5, 15 and 16, inhibitory effect of a-1 on Candida albicans, inhibitory effect of a-2 on Staphylococcus aureus and MRSA, were study at first.Our study indicated that marine fungi from mangrove plant were great resources for new structural metabolites with bioactivities. They are worth further researches.
