| Objective:To investigate the protective effects of jiang tang bao shen decoction (JTBSD) on the diabetic nephropathy in rats and its mechanism.Methods:The Wistar male rats were randomly divided into the normal group and model group.The model group was injected streptozotocin(50mg/kg body weight) intraperitoneally which was dissolved in 0.05mol/l citric acid buffer,pH4.4,4℃.An equal volume of citric acid buffer was injected into the normal group.The fasting blood glucose levels of caudal vein were measured after 72 hours.Animals with blood glucose levels≥16.7mmol/l and urine glucose over +++ were selected as diabetic rats.The diabetic rats were randomly divided into three groups:diabetic model group,JTBSD -treated group and benazepril hydrochloride(BNPH)-treated group.JTBSD -treated group was administrated 30g/kg.d JTBSD crude drug by oral BNPH -treated group was administrated of 10mg/kg.d BNPH suspension by oral; diabetic model group and the normal control group were administrated the equal volume of drinking water by oral,once a day,12 weeks in total.Levels of blood glucose,total cholestrol,blood urea nitrogen,serum creatinine,creatinine clearance rate,urinary protein amounts in 24 hours and kidney hypertrophy were measured 12 weeks after administration;the pathological changes of kidney was observed under light microscope after HE staining;levels of transforming growth factor betal expression were detected with immunohistochemical method.Results:The results showed that the manifestation of polydipsia,polyphagia,polyuric of diabetic model rats were improved significantly in JTBSD group; compared with the model group,the body weight of diabetic rats was increased in JTBSD group,which has a significant difference(P<0.01),the renal weight/body weight,blood glucose,total cholesteroal of diabetic rats were decreased markedly in JTBSD group,which have significant differences(P<0.01,P<0.01,P<0.01); compared with the model group,blood urea nitrogen,serum creatinine,creatinine clearance rate and urinary protein amounts in 24h of diabetic model rats were decreased markedly in JTBSD group,which have significant differences(P<0.01, P<0.01,P<0.01,P<0.01);compared with the model group,the hyperplasia of extracellular matrix,glomerulus hypertrophy,the extension of renal tubule and the edema of epithelial cells were inhibited markedly and the levels of transforming growth factor betal expression was reduced in JTBSD group.Conclusion:JTBSD has obvious protective effects on diabetic nephropathy in rats,its possible mechanism can be contributed to regulating blood glucose and lipid metabolism,inhibiting renal hypertrophy,preventing the change of renal function,reducing the pathological changes of renal rats and inhibiting the levels of cell factor transforming growth factor betal expression. |
