ObjectiveThe article studied on chemical constituents of the fresh radix of Rheum officinale Baill.It need to adress is to study the extraction,separation and purification methods of the natural aldehyde products by a special reaction of sodium bisulfite. The article also investigated the structure-activity relationship of the anthraquinone in the rhubarb.Materials and Methods(1)The correlative researching literature about the rhubarb,including chemical components and pharmacological effects,etc.,have been presented and summarized.We searched the general subjects from both domestic and international literature,including Chemical Abstracts and investigated the structure-activity relationship of the anthraquinone in the rhubarb.After the manufacture of ten batches of semi-works production,the final processing technology have been formulated.(2)From the alcoholic extracts of the fresh plant,by column chromatographies with silica gel and sephadex-18,and identified by NMR,MS,IR,UV spectra.(3)Aldehydes from the fresh radix of Rheum officinale Baill.were isolated by a special reaction of sodium bisulfite and purificated by column chromatographies with silica gel and sephadex-18,,and identified by NMR,MS,IR,UV spectra and other methods.Results(1)From the fresh radix of Rheum officinale Baill.,13 compounds have been isolated. They are three aldehydes:vanillina(XDH-1),parahydroxybenzaldehyde(XDH-2), cinnamyl aldehyde(XDH-3);two stilbenes glucosides:3,5-dihydroxy,4'-anisyl stilbene3-O-β-D glucoside(XDH-4),rhapontin(XDH-5);five anthraquinones: chrysophanol(XDH-6),aloe-emodin(XDH-7),rhein(XDH-8),emodin(XDH-9), physcion(XDH-10);two anthraquinones glucoside:rhapontin 8-O-β-D glucoside(XDH-11),physcion8-O-β-D glucoside(XDH-12);one monosaccharide:α-glucose(XDH-13).Among them,three aldehyde and two stilbenes glucoside were isolated from this plant for the first time.(2)Four aldehydes were isolated and saw from this plant for the first timeby a special reaction of sodium bisulfite.Two,vanillina(XDH-1), parahydroxyben-zaldehyde(XDH-2) were identified by NMR,MS spectra;one, Cinnamic aldehyde(XDH-3) was identified by TLC;because of instability rheinal(XDH-14) is only saw by TCL.Conclusions(1)From the fresh radix of Rheum officinale Baill.,five compound were isolated from this plant for the first time.They are vanillina(XDH-1), parahydroxyben-zaldehyde(XDH-2),cinnamyl aldehyde(XDH-3),3,5-dihydroxy, 4'-anisyl stilbene3-O-β-D glucoside(XDH-4),and rhapontin(XDH-5).The study on chemical constituents supplies the reliable clue to develop the high-level medicine.(2)Four aldehydes were isolated and saw from this plant for the first timeby a special reaction of sodium bisulfite.So,It is a specificity that with sodium bisulfite addition reaction to extract and separate the aldehyde compounds.(3)We review the natural anthraquinone derivative from the Radix et Rhizoma Rhei at the point of the structure-function relationship.The anthraquinone derivative have many identical activities because they have the identical mother nucleus;but the strength of their activities are different,because they have different substituting groups.The anthraquinone derivative appearance evident the structure-function relationship in many respects,such as antioxygenation,antibiosis,anticancer,the influence of immunity and so on.
