Study On The Extraction, Purification And Pharmacological Action Of Total Flavonoids In Sarcandra Glabra

Sarcandra glabra,a sort of common Chinese crude drug,is distributed over the southern of the yangtze river.We can get it with artifical growth.Its chemical constituent,pharmacologic action,clinical application had been researched by many scholars,however,there are seldom researches for flavonoids of Sarcandra glabra.In this paper,we studied the Sarcandra glabra of Guizhou province,not only established the assay method of TFS,but also established technology line of extraction,separation,purification for TFS.Moreover,the effect of hypolipidemic, anti-oxidation and bacteriostatic action for TFS was researched,in order to investigate inhibition effect of heart-cerebrovascular disease.The results showed that:1) Contrasted and improved the tradition assay method of total flavonoids in plant,we established the fitting assay method of TFS—the complexation of Al(NO₃)₃.2) The optimization technology line of water-alcohol extraction for TFS is that: extraction solvent 60%,extraction temperature 80℃;the rate of dosage liquor 1:20; extraction time 3h;extracted 1 time;the extraction rate of TFS is 7.10%.Compared with the tradition technology line,there were the advantadges of lower energy expenditure and minor working procedure in this method.3) The article utilized the method of quiet state adsorption and dynamic state adsorption to study the optimal purification condition of total flavonoids from Sarcandra glabra with macroporous adsorption resins.The results showed that AB-8 macroporous adsorption resins was the best for adsorbing and separating the total flavone in Sarcandra glabra,The optimum extraction conditions were as follows: The concentration and the current velocity of the original solution were 4mg/mL and 2mL/min,the maximum adsorbing capacity for TFS was 8mg/mL,The eluant was 70%ethanol and the eluting velocity was 2mL/min.The consumption of eluant was 3BV.In this condition,the purity of total flavonoids exceed 60%.4) TFS had invariably the effect of kata-bloodfat in experiment mouse. Compared with the model group,the content of TC,LDL-C,TG in blood serum of mouse in every dose group was invariably degraded.5) There are good cleaning effectiveness of DPPH and hydroxyl free radical with the extract of TFS.The EC₅₀ of DPPH and hydroxyl free radical is 2.4mg/mL and 0.73mg/mL.With the same concentration,the cleaning effectiveness of TFS refined extract is more powerful than crude extract's,but,there isn't ad rat between clearance with content of TFS.6) When the concentration of TFS refined extract was 50mg/mL,the bacteriostasis rate to Escherichia coli and Staphylococcus aureus is 47.03%and 27.78%.
