Objective:To investigate the correlation between nestin expression and malignancy within human gliomas.To study the change and significance of nestin expression in differentiation of SWO-38 human glioma cells induced by CDA-2 (Uroacitide),a healthy human urine extraction.Methods:The correlation between nestin expression and the malignant grade has been studied in 56 human gliomas by immunohistochemistry.The change of cell morphology was observed by HE staining with light microscopy and the effect of CDA-2 on GFAP expression was detected by Western blot method in SWO-38 human glioma cells.Expression of nestin in SWO-38 cells was observed by immunofluorescence with confocal microscopy,expression of nestin before and after treated with CDA-2 was detected by RT-PCR and Western blot method.Results:The intensity of nestin immunostaining roughly paralleled the malignancy grade in 56 human gliomas.Light microscopy observation revealed that CDA-2 could induce SWO-38 cells differentiate into astrocyte with increased cytoplasm and cytoplasmic processes as well as decreased nucleus/cytoplasm rate. GFAP expression was upregulated in differentiation of SWO-38 cells induced by CDA-2.Nestin was expressed in cytoplasm and stained like filament by immunofluorescence staining.Nestin expression was downregulated in differentiation of SWO-38 cells induced by CDA-2.Conclusion:The expression intensity of nestin exhibited significant positive correlation with the malignant grade of glioma.CDA-2 upregulated GFAP expression and induced differentiation in SWO-38 cells.Nestin expression was downregulated in differentiation of SWO-38 cells induced by CDA-2 and it may be a new differentiation marker of glioma.
