| AimUsing the hyperlipidemia mouse model,to observe the effect of luffa cylindrica on serum TC,TG,LDL-C,SOD,liver tissue MDA in hyperlipidemia mice,and to synthetically evaluate the hypolipidemic and antioxidant effects of luffa cylindrica.To confirm,by determining the content of polypholic compound in the luffa cylindrica and its antioxidant activity in vitro,that luffa cylindrica contains polypholic compound,also,to isolate the polypholic compound in luffa cylindrica and to perform a preliminary identification.Methods1.The hyperlipidemia model of mice was established,the hypolipidemic effect of luffa cylindrica and its antioxidative activity were observed by using xuezikang as a positive control.2.Acetone soaking procedure was used to isolate the polypholic compound in the luffa cylindrica,Folin-C method was prepared to determine its content,the ability to inhibit the OH·of crude extracts of luffa cylindrica was observed.3.Contents of polyphenols of different breeds and different parts of luffa cylindrica and their abilities of inhibit the DPPH·and OH·were compared.4.Active polypholic compounds were extracted with acetone soaking,organic solvent extraction and column chromatographic procedure.Results1.TC and LDL-C of low-dose group luffa cylindrica,high-dose group luffa cylindrica,were all lower than that of hyperlipidemia model group(P<0.01),among them,the drop of TC and LDL-C of the luffa cylindrica high-dose group was the most obvious;the serum SOD of mice of the low-dose luffa cylindrica group and the high-dose luffa cylindrica group were both higher than that of the hyperlipidemia model group and the control group,among them,the SOD activity of the high-dose luffa cylindrica group was obviously higher than that of the hyperlipidemia model group(P<0.01).2.The content of polyphenols in the fresh luffa cylindrica was 0.3g±0.1g/kg.3.The crude extracts of fresh luffa cylindrica exerted quite a strong capacity to inhibit the creation of the OH·.4.The content of polyphenols in the luffa cylindrica was obviously higher than that in the luffa acutangula(P<0.05).The content of polyphenols in the luffa cylindrica skin was 0.35g±0.1g/Kg,which was 1.7 times of that contained in the flesh of luffa cylindrica.Luffa cylindrica exerted a stronger capacity of inhibiting OH·and DPPH·than luffa acutangula(P<0.01) did.5.The crude extracts of luffa cylindrica were further extracted with acetic ether, the fraction gained exerted a stronger antioxidant effect than the other fractions from other solvent.The fractions from acetic ether were subject to sephadex LH-20 column chromatography for further purifying,the main antioxidation active fraction were fraction D and E,which were determined as flavone by a preliminary identification.Conclusions:1.Luffa cylindrica was able to obviously reduce the lipid level of hyperlipidemia mice and had a distinct antioxidant effect in vitro and in the body.2.Luffa cylindrica was rich in polypholic compound.3.plant polyphenols in Luffa cylindrica were more than that in Luffa acutangula, the content of polyphenols in the skin of luffa cylindrica were significantly higher than that in the flesh of luffa cylindrica,Luffa cylindrica possessed stronger antioxidant ability than luffa acutangula did.4.Via the biochemical technologies such as acetone soaking,acetic ether extraction and column chromatographic procedure,preliminarily purified luffa cylindrica polyphenols were acquired,after a preliminary identification,it was determined as flavone in the polyphenols family,it might be the active component in the luffa cylindrica that contributed to the hypolipidemic and antioxidative effects. |
