Objective To detect the antigen level of plasma plasminogen activator inhibitor-1(PAI-1) and the promoter-675 4G/SG,-844 G→A polymorphism in Chinese Han population,and to investigate whether they are associated with Acute Coronary Syndrome(ACS).Methods 75 patients with ACS(ACS group) and 64 normal controls(control group) were studied.Plasma PAI-1 antigen was measured by ELISA.Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP),denaturing high performance liquid chromatography(DHPLC),and DNA sequence analysis were used to screen the single nucleotide polymorphisms and the genotype distribution of -675 4G/SG and-844 G→A located in PAI-1 gene.Results Genotype frequency of PAI-1 was observed in both groups accords with Hardy-Weinberg.Plasma level of PAI-1 antigen in ACS group was higher than that in control group(67.86±32.59ng/ml vs.51.35±17.95ng/ml,P<0.01) and related to the 4G/5G polymorphism.Carriers of 4G and G alleles have high plasma PAI-1 antigen level.Plasma PAI-1 antigen level was significantly associated with BMI,Glu,TC,LDL.Conclusion PAI-1 plays an important role in regulation of the fibrinolysis function. In patients with ACS the PAI-1 antigen level was much higher.Promoter-675 4G/5G and-844 G→A polymorphism have a relationship with the occurrence of ACS,and carriers of 4G/4G and AG,GG have high plasma PAI-1 antigen level.Elevated plasma PAI-1 activity and BMI,Glu,TC,LDL might be the risk factors of ACS.
