Objective To study the role of tumor necrosis factor -α(TNF-α) and basic fibroblast growth factor ( bFGF) in the onset and development with traumatic acute lung injury(ALI)and their correlationship.Methods 36 health New Zealand rabbits were randomly divided into the normal control group(group C, n = 12) , AL I group (group M , n = 12) and Tumor necrosis factorαmonoclonal antibody(TNFαMAb) treatment group (group T ,n= 12). To set up acute lung traumamodel by weight-dropping. Treatment group were injected TNFαMAb 10mg/kg instantly after acute lung traumamodel. the normal control group and ALI group were injected normal saline instead of TNFαMAb for comparation. 3 rabbits of each group were killed after being extracted blood 1 , 3 ,7 ,and 14 days after acute lung traumamode. One piece of tissue was taken from the injured region and the uninjured region from the normal control group respectively , then HE staining and immunohistochemical staining were performed to measure the TNF-αcontents of blood serum.Results (1)At the 1st day after lung injury, the liquid and cells were exuded into the alveolar antra and alveolar interval of region injured; and at the 3rd day , the exudation increased. But at the 7th day, the exudation decreased,and at the 14th day, which had almost been absorbed. The course of pathological change in the treatment group were less than in the model group respectively. (2) The TNF-αcontents of blood serum was increased from 1stday to 3rd day after ALI(1st day : P < 0.05,3rd day: P < 0.01) , then decreased to normal on 14th day (P > 0.05) . Compared with ALI group, in the treatment group TNFαMAb could decrease the TNF-αcontents of blood serum (1st day : P < 0.05,3rd day: P < 0.01). (3) The bFGF expression increased from 1st day to 3rd day after ALI, and decreased to normal on 14th day. Compared with ALI group, in the treatment group , the bFGF expression decreased respectively.Conclusions (1) After ALI, the TNF-αcontents of blood serum,histopathology and the bFGF expression in injured region change gradually. (2) The correlation between the TNF-αcontents of blood serum and the extent of lung injury is positive. (3) the bFGF is important for improving lung injury. (4) The TNF-αcould be important start-up factor to the bFGF , both could play important action on acute lung injury and repair.(5)TNFαMAb can improve the acute injury lung tissue.
