| Objective To study levels in plasma during the time of pregnancy 10~14weeks, 20~ 24weeks, 30~34weeks and expression in placenta of neurokinin B (NKB) and endothelin-1(ET-1) of patients with hypertensive disorder complicating pregnancy (HDCP) . To investigate the relationship between neurokinin B (NKB) , endothelin-1 (ET-1) and the pathogenesis of hypertensive disorder complicating pregnancy (HDCP).Methods To trace forty-four pregnant women with previously normal blood pressure until after 6 months of deliver. These patients were divided into preeclamptic group (4 severe cases and 6 mild), gestational hypertension group (12 cases) and healthy normal pregnant group (22 cases) according to the follow-up of pregnancy outcome. A prospective study was performed to these forty-four women:(1)Detecting maternal blood plasma level of NKB and ET-1 during the time of early pregnancy(10~14weeks), second pregnancy(20~24weeks)and third trimester(30~34weeks)by method of enzyme linked immunosorbent assay.(2)Exploring expression and distributions of NKB in placenta by method of immunohistochemistry (SP).(3)Studying expression of NKB mRNA and ET-1 mRNA in placenta by method of RT-PCR.Results (1)The levels of NKB and ET-1 could be detected in maternal blood plasma of all 44 subjects at 10~14, 20~24, and 30~34 gestational weeks and zoomed with the development of pregnancy, reaching peak at 30~34gestational week. The levels of NKB and ET-1 in preeclampsia were significantly higher than controls ( P < 0.05). There was no difference between the gestational hypertention placentas and normal one . And we found there were notable positive correlation between plasma NKB and ET-1 levels in preeclampsia ( r= 0.79, P <0. 05) . (2) The distributions of NKB in placentas of HDCP group were similar to normal one, which were in the cytoplasm of villous syneytiotrophoblast, villous vascular endothelial cell and villous stroma, especially in villous syneytiotrophoblast. Expression of NKB were significantly increased in preeclampsia placentas compared to the normal ones (P <0.05). There was no difference between the gestational hypertention placentas and normal one. (3)The transcription levels of the NKB mRNA and ET-1 mRNA in preeclampsia placentas were significantly higher than that of normal groups (P <0.05). There was no difference between the gestational hypertention placentas and normal one .Conclusion: The increased levels of NKB and ET-1 in preeclampsia may influence the development of HDCP together and play a vital role on this disease. |
