| Evaluation of TCM's quality and study on pharmacological action of compound prescriptions are the important subjects to TCM' s modernization.In this paper compound JH-07 was selected as an object for exploring the methodology of TCM's fingerprint and evaulating anti-tumor activities..Through primary pharmacology test,compound JH-07 was proved to have anti-tumor activities.The compound is composed of Rhizoma Coptidis,Fructus Evodiae and Gamboge,which have the effective constituents of berberine,evodiamine,rutacarpine and gambogic acid.Main research work and results were reported us follow:The method of HPLC was adopted in the test to calculate the contents of the effective constituents in plants,in order to solve the content change and unstable pharmacodynamic activity.The contents of berberine,evodiamine,rutacarpine and gambogic acid are 74.5,78.6,46.5 and 26.2mg·g-1, respectively,Fingerprint of the compound by HPLC was established to control the quality of JN-07.The liquid condition of HPLC is:phase system:water-acetonitrile -50 mmol·L-1 KH2PO4(adjust PH to 3 with H3PO4),gradient elution; T=25℃;λ=283nm;velocity:1.0ml·min-1;inject 2OμL.In the chromatographic fingerprint,20 mutual peaks were selected as the fingerprint peaks.Most of the peaks were separated well,with good accuracy,stability and repeatability,satisfied to the requests of fingerprint.The HPLC fingerprint method can be used in the quality control of compound JH-07.In vitro,anti-tumor assay was performed with human esophageal cell EC9706,human leukemia cell K562 sensitive line and drug-resistant line, human fibroblastic cell L929 and human glioma cell TJ-905.MTT method was employed,inhibitory rates and half-inhibitory concentration(IC50) were recorded.IC50 of JH-07 on K562 sensitive line,K562 drug-resistant line and TJ-905 were 15.40μg·mL-1,9.55μg·mL-1,15.02μg·mL-1,respectively.And IC50 of JH-07 on EC9706 and L929 are not satisfied,which shows that compound JH-07 is selective.In vivo,mouse liver cancer H22 was selected and tumor inhibitory percentage,thymus gland,spleen indexes and liver weight were measured. tumor inhibitory rates on H22 sarcoma was 36.5%~48.7%.All JH-07 treated groups compared with control group showed statistical significance(P<0.01). But JN-07 groups showed small differences in spleen index,thymus index and liver weight compared with control group(P>0.05).Nevertheless,when compared with cis platinum group,the differences were with statistical significance(P<0.01).JH-07 had obvious anti-tumor activities,in vitro and in vivo,further work should be done on JH-07. |
