| Objective:To investigate the effects of ghrelin in VMH on gastric motility and its mechanism by which hypothalamic ghrelin is involved in the regulation of gastric motility, and investigate the effects of DVC administration of ghrelin on the immunoreactive expression of c-Fos protein in ARC in rats.Methods: recording of gastric motility in conscious rats and immunohistochemistryResult:(1 ).The amplitude of gastric motility in conscious rats were dose-dependently increased by microinjection of ghrelin into VMH, and the effects were abolished by vagotomy.(2).The amplitude of gastric motility was not increased by microinjection of the GHS-R antagonist(D-Lys~3-GHRP-6) and ghrelin into VMH.(3) DVC injection of ghrelin 10pmol or ghrelin 20pmol can dose-dependently induce the expression of c-Fos protein in ARC.Conclusion:Our results suggest that extro-VMH ghrelin may be involved via GHS-R in the regulation of gastric motility, implying that ghrelin has a functional resemblance to motilin in the central regulation of gastric motility.DVC injection of ghrelin induces the expression of c-Fos protein in ARC, and they are dose-dependent ,which indicates that except ARC, one of the other possible regions where ghrelin exerts its orexigenic effect is DVC. |
