Momordica Charantia L. Var. abbreviate Ser (MCV) is a kind of mutation of Momordica. Charantia L.(MC) which belongs to Cucurbitaceae. It is reported that MCV has been used for the treatment of hypoglycemia. Saponin is one of the main active ingredients in MCV. In this work, the experiments were carried out on the purification of saponins from MCV. in order to provide the reference for the industry production. The research of the consist of the saponins, the method of the hydrolyzation, the comparison of the hypoglycemia between saponins and sapogenins were also carried out in this work.We got the optimum conditions for purification of AB-8 though static and dynamic adsorbent experiments and dynamic elute experiments as following: the elute solvent was ethanol, the concentration of sample was 5-5.5mg·mL-1; the pH of sample is 5; the salty concentration(KCl) of sample was 9mg·mL-1,the flowing speed of sample was 2Bv·h-1. Different concentration of ethanol elute solvents were collected for the sake of the brief purification. 4 times volume resins of 95% ethanol was used to elute the macroporous resins for the sake of getting the crude saponins. The result showed that the purity of saponins was 77.8% and the recovery was 90.1%.A monomer was purified from the ethanol extraction on C18 column and its molecular structure was identified to be cucurbitane-5-en-3β-22,23,24,25-tetrahydroxyl-3-O-3-β-D- glucopyranose (1→6) -D-glucopyranoside by NMR.The studies on sapogenins from MCV. were carried out to study the components of saponins. The method of detection of the sapogenins was confirmed in this work. It was found that we could get higher purity as 75.6% when straightly hydrolyze the saponins. LC/MS analysis showed that three of the compounds sapogenins have the same molecular weight(MW) of C30H48O3, oleananes,β,7β,23-trihydroxy-cucurbita-5,(23E)-dien-19-al which had been reported in MC. We got another sapogenin which has the same MW of the sapogenin fraction of the saponins in later experiment. The rapid detection of sapogenins by TLC showed that the effective develop solvents were chloroform: petroleum ether was 2:8, in which condition we got six bands of sapogenins that matched to the result of LC/MS.The hypoglycemia of saponins and sapogenins from MCV was studied in this article. It was found that saponins could evidently reduce the blood sugar of diabetic rats. The blood sugar of diabetic rats reduced 26.8% after lavage with the dosage of 200mg/mg avoirdupois in 4h and reduced 58.4% in 5d. The sapogenins were not found the function of hypoglycemia in the same experiment conditions.
