It is well known that lung cancer is one of the common malignant tumors and the majority is non-small cell lung cancer(NSCLC),meanwhile Its morbidity and mortality rates are raising year by year.Conventional chemotherapy is applied by the clinician depending on their experience,but,so poor clinical results were got usually because of drug resistance.Therefore,it is necessary that multidrug resistance(MDR) of the NSCLC tissue be detected before operation.Thus,anticancer drugs can be applied to the patients more rationally.However,there are a large number of cancer patients whose tumor tissue is impossibly obtained because of no surgery opportunities,preoperative chemotherapy or other reasons. Therefore,it is important that one of peripheral blood cells be found to detect the expressions of multidrug resistance-associated protein alike that in NSCLC tissue.Objective:To examine the expression of lung resistance-related protein(LRP) and explore its significance in tumor tissue samples,para-carcinoma tissue samples and peripheral blood lymphocytes(PBL) of the patients with NSCLC and normal people,and the relationship among them was analyzed.Methods:Tumor tissue samples and para-carcinoma tissue samples of 66 patients with NSCLC were collected.All specimens were fixed with 10%formalin,and paraffin sections were obtained in pathological department.The S-P immunohistochemical staining (IHC) was used for detecting the expression of LRP in tumors and concomitant para-carcinoma tissues.The relationships between the expression of LRP in tumor tissues and various clinical characteristics were investigated.In addition,before operation,the peripheral blood was obtained from the 66 patients,and peripheral blood of 10 normal people was also collected.PBL were isolated from the peripheral blood by density-gradient centrifugation,and smeared.LRP expression in PBL was determined by immunocytochemical staining(ICC) using a monoclonal anti-LRP antibody.Stastistial analysis was carried out through SPSS 13.0 software.Results:1.The positive rates of LRP expression were 74.2%and 36.4%in tumor tissues and para-carcinoma tissues,respectively.The positive rate of LRP expression in tumor tissues was higher than that in para-carcinoma tissues(P<0.01).No significant difference in expression levels was found among patients with different gender,age,tumor differentiation or histological subtypes(P>0.05).LRP expression was significantly higher in the patients with smoking than those with non-smoking(P<0.05).LRP expression was significantly higher in the patients with lymph node metastasis than those without lymph node metastasis(P<0.01).The positive expression rates of LRP were 100%,88.2%and 50%in T3/T4,T2 and T1,respectively,and there was a significant difference in LRP positive rates among T3/T4,T2 and T1(P<0.01).It was suggested that higher positive expression rates of LRP were found in the advanced NSCLC patients.2.The positive rate of LRP expression in primary tumors was higher than that in PBL with NSCLC(P<0.01).Forthermore,the positive rate of LRP in PBL of the normal people was lower than that of the patients with NSCLC(P<0.01).3.The expression of LRP in tumor tissues was positively correlated with that in PBL of the NSCLC patients(r=0.963,P<0.01).Conclusion:1.LRP expression in NSCLC is associated with tumor growth and lymph node metastasis.2.The LRP levels of PBL may predict the LRP expression of primary tumors in patients with NSCLC.
