Selective Neurodegenrative Effects of H2O2 on Dopaminergic Neurons Objective To investigate the selective neurodegenative effects of H2O2 on the dopaminergic neurons.Methods Using PC12 cells differentiated by nerve growth factor as the model of dopaminergic neurons and Using N2a cells as the control group, each group are expose to H2O2 respectively in vitro, the morphological changes of the cells and abnormal protein aggregation were studied by Laser scanning confocal microimaging system. After intracellular DA depletion with reserpine, the PC12 cell were treated with H2O2, study the protein aggregation and morphological changes.Results After being treated with H2O2 for 16 hours,there were conspicuous ubiquitinatedα-synuclein aggregation in endochylema, the process-1ike of PC12 cell disappeared, the cell body became smaller and smoother and the cell nucleus became bigger. However, there were inconspicuous protein aggregation after N2a cell and PC12 cell which depleted DA by reserpine treated with H2O2.Conclusion H2O2 has the selective neurodegenative effect on the dopaminergic neurons in vitro, to cause ubiquitinated protein aggregation, and it might be closely related to the endogenous reserpine of dopaminergic neurons.PARTⅡThe expression state ofα-synuclein in the peripheral blood of patients with Parkinson diseaseObjective To explore the expression state ofα-synuclein mRNA in the peripheral blood mononuclear cells of Parkinson disease ( PD )and its relationship with PD.Methods 30 PD patients and 30 healthy volunteers were enrolled in the study. Peripheral blood mononuclear cells were centrifugated by lymphocyte separation medium. The mRNA expression ofα-synuclein in peripheral blood mononuclear cell were detected by using reverse transcription and polymerase chain reaction (RT-PCR) technique.Results 1.α-synuclein mRNA was expressed in PBMC of both PD and the controls. 2.The expression state ofα-synuclein showed no significance(P > 0.05) among the all subjects. Conclusion The expression state ofα-synuclein in the peripheral blood mononuclear cells is not associated with PD.
