| Bckgroud: Whether nonviral vector transferred cells could continue expressing effective cytokine in vivo is the vital theme in bioengineering sphere.Object: To study whether TGFβ1 gene transfected into bone marrow stroma could enhance the capability of inducing ectopics osteogensis of BMP-2 derived peptide in vivo.Methods: (K)16GRGDSPC-TGFβ1, a new kind of nonviral local gene delivery system, was manufactured, and was used to transfer TGF-β1 gene to bone marrow derived stroma cell. Then We sieved out the masuline clone, tested it by the SABC method, macrocultured it and combated it with gelatin sponge- BMP-2 drived peptide complex. The complex were implanted into each side of erector muscle of spine of rats: the experimental group at the right and control group at the left. And we studied its capbility of osteogenesis through testing the highest CT image of implanted area by SIENET MagicView 300, activity of ALP and HE dyeing of tissue slice at 3 weeks,5 weeks, and 8 weeks. The control group complex was exactly the same with experimental group except without TGFβ1 gene.Result: The experimental group showed masulin in TGFβ1 dying by SABC method. Ectopic osteogenesis was found in both groups, and it's time dependence could also be observed. The highest CT values were repectively 254±20,331±34 and 477±37 in expermental group, which were obviously higher than those of control group, whose CT values were respevtively 237±17,298±31,433±45. Activity of ALP was respectively 57.78±0.64,65.7±0.59,71.83±0.67 in experimental group, and was 55.46±0.55, 63.27±0.53,70.32±0.63 in controling group. And HE dyeing of tissue slice demonstrated more bone formation in experimental group.conclusion: TGFβ1 gene transfected bone marrow stroma cells by oligopepitide (K)16GRGDSPC combated with gelatin sponge could enhance the capability of inducing in vivo ectopic osteogenesis of BMP-2drived peptide. |
