Objective:To investigate the expression ofα1A-adrenergic receptor(α1A-AR) and muscarinic acetylcholine receptor M1(mAChRM1) in liver tissue of fibrosis in mouse infected with Schistosoma japonicum and the possible mechanism of the neurotransmitter and its receptor in liver fibrogenesis.Methods:Thirty mice were randomly divided into two groups, group N was a normal group(10) and group M was a model group(20). Mouse was infected with Schistosoma japonicum by transfering of the cercaria to its abdominal space in the model group. After twelve weeks, the livers of all mice were collected. The pathological changes of liver were observed by light microscope and electron microscope. We detected the expression ofα1A-AR and mAChRM1 by immunohisto-chemistry,RT-PCR and Western-blot in the normal livers and the model livers that infected with Schistosoma japonicum.Results:The morphological examinations showed typical pathological changes in liver fibrosis with worm tubercles of schistosoma deposited in the liver tissue, fibrosis around the worm tubercles and infiltration or soakage of inflammatory cells, the immunohistochemistry showed that in normal liver there was separated expression ofα1A-AR and mAChRM1 in enchylema of hepatocytes, these expressions increased markedly in model liver infected with Schistosoma japonicum. The results of RT-PCR and Western-blot also showed the same results that the expression increased in model group compared with the normal group, the difference was significant in statistics(P<0.05).Conclusion:The expression ofα1A-AR and mAChRM1 significantly increased in hepatic fibrosis in mouse infected with Schistosoma japonicum. So we conclude that the neurotransmitter and its receptor may play a role in liver fibrogenesis. Keywords:...
