| Aim:Based on the optimized results of single-factor,taking the content of Atractylodes I in fired Atractylodes macrocephala Koidz,which was determined by RP-HPLC,as an index,the optimal supercritical fluid extraction technological parameters were obtained by orthogonal experiment;The liposoluble components of fired Atractylodes macrocephala Koidz were analysed by GC/MS;The effect of Atractylodes liposoluble components on small intestine movement function of mice was investigated,and its possible mechanism was explored.Methods:Factors influencing the extraction yield were examined through orthogonal experiment:such as extracting pressure,extracting temperature,separation pressure,separation temperature,extraction time,flux of CO2,particle size of Atractylodes,etc.the optimal supercritical fluid extraction technology were obtained, according to the yield of Atractylodes I under various experimental conditions.The liposoluble components of fired Atractylodes macrocephala Koidz extracted by soxhlet extraction method and supercritical fluid extraction method,were analysed and compared by GC/MS;the experiment of intestinal propelling and gastric emptying were carried in normal mice and mice pretreated with atropine,the effect of Atractylodes Macrocephalakoidz on gastrointestinal function were observed at the dosages of 25,50, 100 mg/kg.Results:Base on the result of orthogonal experiment,the optimum supercritical fluid extraction process was as follows:extracting pressure 35MPa,extracting temperature 50℃,separation pressureâ… 8MPa,separation temperature 30℃,extraction time 2.0h,flux of CO2 30kg/h,particle size of Atractylodes 60 mesh;The extraction rates of liposoluble components with soxhlet extraction was 97.206%,the components examined,32 of the 37 were identified.The extraction rates of liposoluble components with SFE-CO2 was 80.451%,the components examined,24 of the 58 were identified; Atractylodes Macrocephalakoidz had obviously promotion effects on enterokinesia and gastric emptying both in normal mice and pretreated mouse.Conclusion:Under the optimal extraction conditions,the yield of Atractylodesâ… in fired Atractylodes macrocephala Koidz was higher.The liposoluble components of Atractylodes macrocephala Koidz with different extraction methods have significant deviation,the extraction rates is higher and the components are more with the method of SFE-CO2。the CO2 supercritical extraction products of fired Atractylodes macrocephala Koidz can promote stomach movement. |
