| Hepatitis A and hepatitis B are the most popular virals hepatitis disease and infection disease that is harm to public health.The virals infection of hepatitis B can develope chronic liver disease,liver cirrhosis and liver cancer.Usually,the virals infection of hepatitis A can not develope into chronic liver disease,minorities of adult patient's condition can be protracted and recurrented.The course of individual patients' is reach to six months and prognosis is worse.Because hepatitis A and hepatitis B are awfully harmful to human public health,and there is still not a specific drug treating these two virals diseases,the generally utilization of HAV vaccine and HBV vaccine can contain these two virals diseases.Now,for protecting two or multiple diseases, advanced development and utilization combined vaccine including two or mulitple antigens is paid attention to by every countries in the world,and combined vaccine is become into hot spot in the field of vaccine research.Utilization combined vaccine is necessary for simlify and expansion of children vaccination planning.Combined hepatities A and B vaccine is a combined vaccine that possess double specific protection function to aim directly to HAV and HBV.In the research of combined hepatities A and B vaccine,researchers found that combined vaccine can induce higher antibody than univalent vaccine and generally consided that combined vaccine can induce higher HBV antibody than univalent HBV vaccine,and presume that HAV and HBV antigen may interact each other as a adjuvant,but there is still no systemic immunology report.We tested four indexs of 150 mice immuned by two different concentrations of combined HAB vaccine,univalent hepatitis vaccine,we use flow cytometry to detect cytokine and T cell subset typing,use MTT assay to detect spleen cell proliferation and ELISA assay to detect antibody,and then evaluate cell and humoral immunity response of combined vaccine,judge the character of immune response,evaluate the effect of immunologic memory,and find the best matching between hepatitis A and hepatitis B antigen,and analysis the two antigen's interaction mechanism in combined HAB vaccine and know the immunogenicity of combined HAB vaccine wholly.We adopted HAV inactivated vaccine produced by Medical and Biology institute and gene engineering recombination HBV vaccine produced by yeast half-finished product,mix 640Eu/ml HAV antigen into 10ug and 20ug HBV antigen.200μl different contribution combined vaccine or univalent hepatitis vaccine were injected into gastrocnemius muscle.150 female BALB/C mice were divided in 6 groups randomly, 25 mice/group,were injected 1mg/ml,640Eu/ml,10μg/ml,20μg/ml, 640Eu/ml:10μg/ml,640Eu/ml:20μg/ml in control group,univalent hepatitis A, univalen hepatitis B,univalent hepatitis B,combined HAB vaccine,combined HAB vaccine.We choose prime-boost strategy one time on the forth week.The blood in fossa orbitalis and spleen cell were collected on the first,second,third,fourth,sixth week after the first injecting from 5 in every group.the indexs represent humor and cell immunity were detected.The result indicated that two combined vaccine can stimulate more stronger immune response than univalent vaccine.In the earily period of the first immunization,HAV antigen can boost HBV vaccine cell immunogenicity.We aggregate analysis the result and conclusion is that combined HAB vaccine manufactured independence by medical and biological institute have the good immunogenicity,combined HAB vaccine can boost vaccine specific immune response on the initial and boost immunity and induce Th1/Th2 cytokine that is higher or same than univalent hepatitis A vaccine and univalent hepatitis B vaccine after the boost immunization. |
