Study On The Total Synthesis Of Saucernetin-8

Saucernetin-8,structurally related and functionally unique dineolignans,has been isolated from active root extracts of Saururus cernuus L.Recently Saucernetin-8 has been shown to be a potent inhibitor of the key transcription factor HIF-1.In this thesis, full details are showed for the total synthesis of Saucernetin-8 by using 4-pentenoic acid as starting material,followed by the use of Evans Reagent,asymmetry Aldol reaction,through TBS protection,removing of the oxazolidinone,LAH reduction,NaIO₄ oxidation,Weinreb ketone synthesis reaction,ect...Saucernetin-8 was synthesized successfully in 21 steps.By using of the two intermidiates of the route,we got two isomers of saucernetin-8.The molecular structure of them have been confirmed by ¹H NMR,¹³C NMR,HPLC-MS.
