| Objective:To investigate the association between the single nucleotide polymorphism(SNP) of two kinds of genes, called Extracellular Superoxide Dismutase(EC-SOD) and adiponectin respectively, and the development of Typeâ…¡Diabetes Mellitus(T2DM) in the Han population in Jilin region.Methods: This original research was divided into research objects by two sets. The first group was patients of T2DM,including 105 persons(57 male, 48 female, and average age was 56±11.4); The second group was people of Normal control(NC), including 49 persons(29male, 20 female and average age was 47士8.44). These two sets were arranged for a contrast control study. First quick extraction of the human genome DNA was carried out from leukocytes of people's peripheral blood with a chemical reagent,KI.Secondly,The Ala40Thr polymorphism of EC-SOD gene was verified by Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) analysis while the I164T polymorphism of adiponectin was observed by Polymerase Chain Reaction-Single Strand Conformation Polymorphism (PCR-SSCP) analysis.Among the process,An electrophoresis with an agarose concentration of 2% was performed,and fluorescent bands were observed under ultraviolet light in order to check whether the process of PCR was successful.For EC-SOD gene,successful PCR products were cut by an endonucleases,named Pauâ… so that every genotype can be determined.For adiponectin gene, a 8% non-denaturing polyacrylamide gel electrophoresis was performed with the single strand DNA from PCR products,then the bands can be checked if there was a migration of single strand between the two groups by silver staining. Comparation of genotypic frequency and allele frequency between these two groups was made with statistic check ofχ~2 .Results: (1)According to Hardy-Weinberg's law of genetic equilibrium,genotype frequency of two groups has achevied a status of genetic balance(group T2DM:χ~2=0.344, P=0.558>0.05 and group NC:χ~2=1.180,P=0.277>0.05) and they both have a community representation.(2) SNP of the EC-SOD gene(Ala40Thr):The genotypic frequency of group T2DM was significantly higher than that of group NC (χ~2=6.909,P=0.032<0.05); allele frequency of group T2DM was also significantly higher than group NC(χ~2=5.198,P=0.023<0.05).(3) SNP of the adiponectin gene(I164T): there were no obvious bandmigrations of single DNA strands between group T2DM and NC samples.Conclusions: (1)Each group has fulfilled Hardy-Weinberg's law of genetic equilibrium and hence has a community representative.(2)According to present samples,the Ala40Thr polymorphism of EC-SOD gene(G→A) may have associations with the development of T2DM in Jilin region and A allele may be a risk factor of development of T2DM.(3)According to present samples,the I164T polymorphism of adiponectin gene(T→C) may be not related to the development of T2DM and C allele may not be a risk factor of development of T2DM. |
