Objective:With the research of tumor developing mechanism,epigenetic modification has become a new view.To study the pathogenesis of cervical cancer in the level of histone modification from the epigenetic modification,we evaluate the level of histone H3 lysine 9 trimethylation in cervical squamous carcinoma tissue.Method:We collected the study groups from 5 patients' tissues with squamous carcinomas of the cervix and the control group from other 5 patients' with normal cervical tuisse.Here we adopted Chromatin immunoprecipitation linked to microarrays(ChIP-chip),for the first time,to profile the variations in H3K9me3 in CpG island regions in squamous carcinomas of the cervix patients and controls' ChIP-qPCR is used to validate the microarray results.Using quantitative reverse transcription-polymerase chain reaction(qRT-PCR) detect the mRNA expression of MBD4,PCGF2,CD44 which have significant difference in the level of H3K9me3 to explore the relationship between the level of H3K9me3 and gene expression.Result:The methylation of 60 genes display increased and 110 genes display decreased which are displayed significant H3K9me3 differences were found in tumor group comparied with normal group,respectively.The results of ChIP-qPCR were coincided well with microarray.Using qRT-PCR we detect the mRNA expression levels of MBD4,PCGF2,CD44 gene.Compared with the control,the mRNA levels of MBD4 and PCGF2 genes in cervical squamous carcinoma are 0.02±0.01 times,0.04±0.03 times lower.The H3K9me3 levels of MBD4 and PCGF2 genes are 2.16±0.01 times,1.36±0.03 times higher.The mRNA levels of CD44 genes in cervical squamous carcinoma are 8.63±0.04 times higher. The H3K9me3 levels of CD44 gene are 0.714±0.04 times lower.Conclusion:There are significant differences in H3K9me3 profiling between tumor group and normal group,which expression is up-regulated in 60 genes,down-regulated in 110 genes.The high level of H3K9me3 can inhibit the expression of related gene,conversely,the low level of H3K9me3 can promote the expression of related gene.CD44,MBD4,PCGF2 exist abnormal expression in cervical cancer.
