Object:In recent year,a large number of researches showed that the incidence of tumor development has related to the disorders of cell cycle.CyclinD1 is one of the most important gene in the cell cycle regulation system.The disorder is playing a crucial role for the formation of turmor.Caveolin-1 exists in a variety of cell surface. It takes part in regulating cell cycle and signal transduction.We took real-time fluorescent quantitative PCR(FQ-PCR) to detect the expression level of caveolin-1 mRNA and cyclinD1mRNA and analyze the correlation between the two genes.The aim of our study was whether the two target genes could be used on the clinical paitients as a new tumor marker,improving the diagnosis in colorectal cancer.It would provide the evidences for the diagnosis,therapy,prognosis at the early stage of tumor and for the target gene of the treatment for colorectal cancer.Method:We detect the expression levels of caveolin-1 and cyclinD1 between 30 colorectal carcinoma tissue after the resection and 12 normal mucosa tissue that were 10cm away from the edge of the tumor by real-time fluorescent quantitative PCR (FQ-PCR).We choose the housekeeping geneβ-actin and analyze gene expression data using the 2-△△ct method.It is a convenient way to calculate the relative changes in gene expression from real-time fluorescent quantitative PCR experiments.Make use of spss 13.0 stat software to deal with and analyze the expression of the two genes and the relativity between the clinical pathology data.Results:The relative expression of caveolin-1 mRNA is 0.66±0.25 in the 30 colorectal carcinoma patients.Caveolin-1 mRNA is down-regulated by 0.66 times in colorectal carcinoma than that in normal colorectal tissue.Meanwhile,the relative expression of cyclinD1 mRNA is 5.94±2.67 in colorectal carcinoma patients and up-regulated about 5.94 times.Caveolin-1 and cyclinD1 are significantly different in differentiated grade(t=2.281,-2.164,p<0.05) and Lymph node metastasis(t=-2.272, 2.219,p<0.05).The expression of caveolin-1 has no correlation with Dukes stages (F=1.895,p>0.05).In contrary,the expression of cyclinD1 has correlation with Dukes stages(F=4.171,p<0.05).There are no relation to the expression of caveolin-1 and cyclinD1 in sex(p>0.05),age(p>0.05),and tumor position(p>0.05).Conclusion1,Compared with normal tissue,caveolin-1 gene is down-regulation and cyclinD1 gene is up-regulation in colorectal carinoma.There are a negative correlation of them.2,The expression levels of the two genes in colorectal carcinoma patients are remarkably related to differentiation grade and Lymph node metastasis.3,The disorders of two genes in colorectal tumor can coexist and participate in negative regulation of tumor occurrence and development.4,The expression levels of the two genes might be seen as the tumor markers in the colorectal carinoma.
