Partâ… The study of tumor formation of paratumor cirrhosis liver cellObjective:To investigate the tumor formation of paratumor cirrhosis liver cell line QSG-7701 hypodermic nude mice influenced by aflatoxin B1 and phenobarbital,and explore the relationship between paratumor cirrhosis liver and liver neoplasm and study the pathogenesis of liver neoplasm.Methods:The present study was designed to culture cell lines(the normal liver cell line HL-7702,the paratumor cirrhosis liver cell line QSG-7701 and the HCC cell line QGY-7703) in vitro to hypodermic nude mice,and injected aflatoxin B1(AFB1),phenobarbital(PB) in HL-7702 and QSG-7701 groups, contrasted by sodium chloride,observeing the tumor formation of three cell lines.Results:There were no tumor formation in pure HL-7702 group and HL-7702 PB group.The tumor formation in HL-7702AFB1 group was 50%,and HL-7702AFB1+PB group was 66.67%.No tumor formation in pure QSG-7701 group,but in QSG-7701AFB1 group(60%),QSG-7701PB group(66.67%),QSG-7701AFB+PB group(80%) and QGY-7703 group(80%),there were tumor formations.Conclusions:Paratumor cirrhosis liver cell QSG-7701 had more features of precancerous lesion.QSG-7701 is an essentially different group from normal liver cell line HL-7702 and HCC cell line QGY-7703,which demonstrated paratumor cirrhosis liver cells were belonged to transforming precancerous cells.It implied paratumor cirrhosis might a key stage in malignant transformation of hepatic cells. Partâ…¡The expression and significance of DLK1 and AFP in nude mice transplanted tumor and hepatocellular carcinomaObjective:To study of the expression and significance of DLK1 and AFP protein in nude mice transplanted tumor and human hepatocellular carcinoma.Methods:Detected the expression of AFP and DLK1 in nude mice transplanted tumor of the normal liver cell line HL-7702,the paratumor cirrhosis liver cell line QSG-7701 and the HCC cell line QGY-7703 by immunohistochemical staining.The expression of AFP and DLK1 in 10 case human normal liver,22 cases paratumor cirrhosis liver and 22 cases hepatocellular carcinoma of human tissue were also detected by IHC.Results:There were no AFP and DLK1 expression in normal liver cell line HL-7702AFB1 group,one case in HL-7702AFB1+PB group expressed AFP and DLK1.No AFP and DLK1 expressed in paratumor cirrhosis liver cell line QSG-7701 AFB1group,one case in QSG-7701PB group expressed AFP and DLK1,2 cases in QSG-7701AFB1+PB group expressed AFP and DLK1.In QGY-7703 group,there were 3 cases expressed AFP,4 cases expressed DLK1.DLK1 was positive in human paratumor cirrhosis cell and the positive rate was 27.27%.The expression rate of DLK1 in HCC tissue was 54.55%,higer than AFP(40.9%);and in AFP negative HCC tissue,the expression rate of DLK1 was 22.73%. Conclusions:1.There were both AFP and DLK expersssion in the nude mice transplanted tumors.2.It was a novel finding that DLK1 expersssion was positive in the human paratumor cirrhosis cell,and positive rate was 27.27%.3.Combination of DLK1 and AFP detection may raise the detection rate of human hepatocellular carcinoma,which have essential diagnostic significance in hepatocellular carcinoma,and DLK1 may be a new marker in early diagnosis of hepatocellular carcinoma.
