Objective: To study the effects of the expression of Runx3, Sp1 and Bcl-2 on carcinogenesis and progression in gastric adenocarcinoma, and provide experimental evidence for prognosis and biotherapy.Methods: The expression levels of Runx3, Sp1 and Bcl-2 were measured by SP immunohistochemistry in 63 cases of gastric adenocarcinoma, 19 cases of low-level intraepithelial neoplasia tissues and 10 cases of normal gastric mucosa. Their relationship relating with clinic-pathological features and survival was also analyzed.Results: (1) The positive rates of Runx3, Sp1 and Bcl-2 in gastric adenocarcinoma were 39.7%,58.7% and 65.1% respectively, 39.7%,58.7% and 65.1% respectively in low-level intraepithelial neoplasia tissues, and 90%, 10%, 20% respectively in normal gastric mucosa(Runx3:P<0.01;Sp1:P<0.01;Bcl-2:P<0.05). (2) The positive expression of Runx3 was correlated to the differentiation, invasion depth and tumor node metastasis of cancer cell (P<0.05). The positive expression of Sp1 was correlated to the invasive depth, tumor node metastasis and clinical stages of cancer cell (P<0.05), but had no correlation to the differentiation of tumor. The positive rate of Bcl-2 was correlated to the differentiation of cancer cell (P<0.05), but had no correlation to the invasive depth, tumor node metastasis and clinical stages of tumor. (3) Spearmen rank correlation showed a negtive correlation was observed between Runx3 and Bcl-2 (P<0.01), a positive correlation between Sp1 and Bcl-2 (P<0.01), and no correlation between and Runx3 and Sp1. (4) The Kaplan-Meier survival analysis showed that the survival rate of positive Runx3 was better than that of the negative groups.The survival rate of positive Sp1 was lower than that of the negative groups. There was no statistically difference between the positive and negative groups of Bcl-2. Conclutions: The results showed that the lower expression level of Runx3 and high expression level of Sp1 were associated with the progression of gastric adenocarcinoma while high expression level of Bcl-2 might act in synergy.
