Objective: To observe the HPSE expression after transiently transfecting human colorectal carcinoma cell HT-29 using siRNA, to approach the effect of RNAi targeting HPSE on the proliferation,invasion and metastasis of colorectal carcinoma cell.Methods: The heparanase mRNA-targeted double-standed siRNA was designed with the bioinformatics technology. For 72 hours after transfecting HT-29 cells with specific anti-HPSE siRNA or N.C.FAM siRNA, MTT method was used to detect the proliferation of the cells every day in three days after transfection. The level of HPSE mRNA in the cells was measured by real time PCR, immunnohistochemistry was taken to detect the expression of HPSE protein, the invasiveness of HT-29 cell in vitro was measured quantitatively by the invasion test of Transwell chamber.Results: Compared with control groups, cell proliferation of siRNA group was obviously suppressed(P<0.01) , The expression of HPSE mRNA in the cell of siRNA group were reduced by 73.04%(P<0.01),but control groups had no obvious difference (P >0.05).Conclusions: (1)Silencing HPSE could effectively inhibit the proliferation and invasion of human colorectal carcinoma. (2)Silencing HPSE, the changes of the cell's biological activity are correlated with the downregulation of level of HPSE mRNA and protein.
