Objective: Study the expression of Maspin,uPA and LN-R in Breast Cancer,and to explore their relationship with clinic-opathologic factors and the correlation of the three factors.Methods: The expression of Maspin,uPA and LN-R in 60 cases of invasive ductal carcinoma of breast,30 samples of tumor adjacent normal breast tissue and 40 metastatic lymph nodes were examined by immunohistochemical staining with Mouse anti- Maspin monoclonal antibody,Mouse anti- uPA multiclonal antibody and Mouse anti-LN-R monoclonal antibody respectively.Results: The positive expression of Maspin,uPA and LN-R in 60 cases of invasive ductal carcinoma of breast was 46.7%(28/60),61.7%(37/60)and 68.3%(41/60)respectively,showing a significant difference with 30 samples adjacent normhal breast tissue which was 76.7%(23/30),30.0%(9/30)and 36.7%(11/30)(P<0.05). The positive expression of Maspin,uPA and LN-R in 30 cases of invasive ductal carcinoma of breast which had metastatic lymph nodes was 26.7%(8/30),36.7%(11/30)and 86.7%(26/30)respectively,and in the 40 metastatic lymph nodes were 5.0%(2/40),65.0%(26/40)and 100.0%(40/40),the differences had statistic significance (P<0.05). The expression of Maspin was significantly correlated with the lymphatic metastasis, surgical-pathologic staging and ER(P<0.05),and had no significance with age,tumor size, histological grade and PR (P>0.05). The expression of uPA was not correlated with age,tumor size(P>0.05),but correlated with lymphatic metastasis,surgical-pathologic staging, histological grade,ER and PR(P<0.05). While the expression of LN-R was significantly correlated with the lymphatic metastasis, surgical-pathologic staging and histological grade (P<0.05),it had no correlation with age,tumor size, ER and PR (P>0.05).Among the 60 cases,12(20%) showed Maspin and uPA positive expression simultaneously ,30(50%) showed LN-R and uPA positive expression simultaneously,and 12(20%)showed Maspin and LN-R positive expression simultaneously. Expression of Maspin was negatively correlated with expression of uPA(r=-0.362,P<0.05); Expression of LN-R was positively correlated with expression of uPA(r=0.348,P<0.01); Expression of Maspin was negatively correlated with expression of LN-R(r=-0.512,P<0.05).Conclusions: There are overexpresions of uPA and LN-R in breast cancer but down regulated expression of Maspin. The carcinogenesis,invasion and metastasis of invasive ductal carcinoma of breast may be correlated with the abnormal expressions of Maspin ,uPA and LN-R .They could be valuable tumor markers and prognostic factors for invasive ductal carcinoma and could be potential target for therapies of invasive ductal carcinoma. The biological characteristics of breast cancer and their lymph-nodemetastatic tumors should be considered together for estimating malignant degree of cancer cell,chosing treatment prescription and predicting the prognosis of patients.
