| Backgroud:The Flos Caryophylli is the dry bug of Eugenia caryophyllata Thunb which is remained with mytle family.Clove's acrid is flavour,warm in prooerty,to the spleen,stomach,kidney.Lilac is the drugs for dispelling cold,warm the body and dispelling cold,reinforce the kidney yang,warm blood.The clove can therapy deficiency cold in spleen and stomach,hiccup, vomiting and diarrhea,bosom spasmodic colic,kidney deficiency and sexual impotence.Because Clove'gas fragrant and is dood at walking,it can up to heart and lung,"treatment chest distress"(《Compile of Materia Medica》), down to liver and kidney could be "kidney treatment,bentun gas,yin pain, strengthing yang and warm waist and knee".Dingyusishen pulvis which is composed of clove and curcuma longa can cure coronary heart disease.This prescription can ease angina pectoris and ameliorate electrocardiogram and so on.Eugenol is the main component of clove volatile oil,up 80.33 percent of its content.Eugenol can be against the Ca2+ catalytic reaction by non-competitive,inhibit the formation of hydroxyl radicals,protect cell membrane lipids against oxidation.Eugenol can clearance the hydroxyl radical of Fenton reaction significantly,and its specific activity higher than the hydroxyl radical scavenger mannitol,on riboflavin methionine light generated active oxygen reduction is also clear the role can be used as exogenous the use of oxygen scavenger.A Japanese study found that eugenol can inhibit the activity of xanthine oxidase,and superoxide dismutase inhibited the chemical produced,so that the direct capture of its superoxide dismutase.Eugenol consider this issue by removing the possibility of myocardial hypoxia/reoxygenat injury and oxygen free radicals produced by damaged protective role of myocardial cells,it is worth further study. Aim:The myocardial hypoxia/reoxygenat(H/R) injury model was set up with Na2S2O4 and vestigate the influence of eugenol against H/R in cultured cardiac myocytes.Methods:Primary culturing baby rat cardiomyocytes,and establishing the hypoxia/reoxygenation injury model by sodium dithionite(Na2S2O4),using the inverted microscope morphological changes of myocardial cells,tetrazolium salt(MTT) Determination of cell survival,flow cytometry cell mitochondrial membrane potential(MMP),and determination of its cell culture medium of lactate dehydrogenase(LDH),malondialdehyde(MDA),superoxide dismutase(SOD) vitality,in order to observe the eugenol on hypoxia/reoxygenation injury in myocardial cells.Results:After being established by Na2S2O4,Myocardial cells' survival rate, MMP,SOD decreased,LDH,MDA increase.Model group,cell survival rate,LDH compared with the normal group,there were significant differences of statistical significance(P<0.001),model group cell MMP,MDA,SOD activity compared with the normal group,there was no significant difference(P>0.05); Compared with model group,eugenol significantly increased hypoxia/ reoxygenation injury of myocardial cells survival rate,there is some tendency of reduction in LDH,MDA and enhancing SOD,MMP.Conclusion:Eugenol can significantly improve Na2S2O4 caused by hypoxia/ reoxygenation injury in the survival rate of cardiac myocytes to a certain extent,reduce the damage of myocardial cells and LDH leakage of MDA content, SOD activity and to increase the stability of the trend of MMP myocardial cells,Eugenol has a certain protective effect of the hypoxia/reoxygenation injury by Na2S2O4 of the cultured in vitro rat cardiac myocytes... |
