Objective: To put forward a large-scale genotype analysis with matrix-assisted laser desorption / ionization time-of-flight mass spectrometry, MALDI-TOF-MS, and to probe into the relationship between DNA repair gene single nucleotide polymorphism and the non-Hodgkin lymphoma heredity susceptivity.Methods: The MassARRAY method was applied to detect the DNA repair gene single nucleotide polymorphism in 73 cases of NHL and 540 cases of normal healthy controls. Meanwhile, the relationship between different genes and lymphoma risk, and the relationship between case classification and prognosis are compared.Result: The distribution of the gene allele frequencies of XRCC1 R280H and MGMT L84F have statistical significance in case of illness and the control group, P=0.01 and P= 0.036 respectively. The frequencies of XRCC1 R280H and MGMT L84F with at least a mutant gene have different distribution between the NHL cases and the controls, compared with the wild homozygote genotype (P=0.000, P=0.029). While, for the diffuse large B- cell lymphoma (DLBCL) of the B-NHL, only the distribution of the XRCC1 R280H gene is statistically significant between patients and controls (AG+AA/GG, P=0.003,OR=0.32, 95%CI=0.145-0.706). From the statistic data, XRCC1 R280H may decrease the risk of NHL development (OR=0.309, 95%CI=0.168-0.567). However, the risk factor of NHL may be the gene MGMT L84F (OR=2.085, 95%CI=1.069-4.068). For the DLBCL, XRCC1 R280H gene reduce the risk of the disease development (OR=0.32, 95%CI=0.145-0.706). By the Kaplan-Meier and log-rank, we found no statistical difference between the patient of having at least a mutant gene and wild homozygote in MGMT L84 and XRCC1 R280H (P>0.05).Conclusion: DNA repair genes XRCC1 R280H and MGMT L84F possess significant correlation with NHL in polymorphism.
