Objective To investigate the relationship between the expression of visfatin in adipose and placenta tissue and glycometabolism in gestational diabetes mellitus(GDM).Methods The expression of visfatin mRNA and protein were separately determined by reverse transcription polymerase chain reaction(RT-PCR) and Westin blot from 40 pregnant women with GDM(GDM group)and 40 pregnant women with normal glucose tolerance (control group).The levels of serum visfatin,fasting plasma glucose(FBG) and fasting insulin(FIN) were measured severally by Enzyme-linked immunosorbent assay(ELISA), glucose oxidase,radioimmunity assay.Insulin resistance index(HOMA-IR) was calculated.Results(1) The serum levels of visfatin,FBG,FIN and HOMA-IR were significantly higher in GDM group(73.1±11.9 ng/ml,4.52±0.46 mmol/L,10.61±3.09 mU/L,2.44±0.93)than those in the control group(35.8±13.8 ng/ml,4.12±0.38 mmol/L,7.42±2.87 mU/L,1.32±0.56)(P<0.011).(2)The levels of visfatin mRNA and protein expression in visceral adipose tissue(0.96±0.13,0.95±0.12) in GDM group were significantly higher than those in the control group(0.56±0.10,0.58±0.13)(P<0.01).(3) The levels of visfatin mRNA and visfatin protein expression in placenta tissue in GDM group(0.69±0.10,0.70±0.11) were significantly higher than those in the control group (0.54±0.09,0.57±0.12)(P<0.01).(4) Correlations was found between serum visfatin levels and the levels of visfatin mRNA and protein expression in visceral adipose tissue,HOMA-IR in GDM group(r==0.799,0.803及0.792,0.794)(P<0.01),but there were no correlations between placenta tissue and serum visfatin and HOMA-IR (P>0.05)Conclusions Increased serum levels of visfatin result in overexpression in visceral adipose,which may be involved in the regulation of glycometabolism in women with GDM.
