Objective This study was designed to evaluate the protective role of Huoxuehuayu prescription on rat's retina injury reduced by acute high intraocular pressure and to observe the expression changes of BDNF with the description.Methods one hundred wistar rats were randomly divided into tow groups A and B.In A group,50 wistar rats' retina were H-E stained and immune-fluorescent(IF) ostained and observed under light microscope;In B group,BDNFmRNA in 50 wistar rats' retina were detected by Real-Time quantitative reverse transcription polymerase chain reaction, (Real-Time qRT-PCR analysis).All rats in A and B group were randomly divided into ten groups.For example,in A group,A0 standed for normal group,A'standed for puncturing group,in which the rats were only punctured but not pressurized,A1~A4 standed for pressurizing groups,in which the rats were sacrificed at 1d,3d,7,14d respcectively after pressurizing,A5~A8 standed for pressurizing and pouring medicine groups,in which the rats were poured with Huoxuehuayu prescription immediately after pressurizing and were sacrificed at 1d,3d,7,14d respcectively after pouring drugs.The method of subgroup in B group was the same as A group.Results1.Histological examination:1d after pressurizing,full-thickness of retia were dropsical severely,the number of RGCs was decreasing,and there were cellules appeared around the nucleus,the thickness of retia was increasing;3d after pressurizing,the dropsical condition was more alleviative than that of 1d,the nucleus of RGCs were swollen further;7d after pressurizing,the dropsical condition was disappeared generally,the number of RGCs continued to descrease,the thickness of retia was thinning; 14d after pressurizing,all constructions of the retina were disordered,we can rarely see the RGCs,the shape of the nucleus became abnormal and karyorrhexis occurred.The changes of retinal shape and the degree of the injury in the pressurizing and pouring medicine groups were less severe than the pressurizing groups during the same time stage.2.IF results:1d after pressurizing,the protein expression of BDNF in GCL and INL was increasing obviously and got to the peak(P<0.01);3d after pressurizing,the protein expression of BDNF started descreasing but was more than the normal group(P<0.05); 7d after pressurizing,the protein expression of BDNF was continued to descrease;14d after pressurizing,the fluorescent particles of BDNF were less than any groups,and the fluorescence was extremely weak(P<0.01).The protein expressions of BDNF in the pressurizing and pouring medicine groups were more than those in the pressurizing groups with the same time stage.(P<0.05).3.Real-Time qRT-PCR results:In pressurizing groups,the expressions of BDNFmRNA started to increase on 1d after pressurizing and got to the peak(P<0.01 ),3d after pressurizing,the expressions of BDNFmRNA started to decline,but were more than that in normal group(P<0.05),14d after pressurizing,it declined at a low ebb(P<0.01).The expressions of BDNFmRNA in the pressurizing and pouring medicine groups were more than those in the pressurizing groups with the same time stage.(P<0.05).Conclusion The Huoxuehuayu prescription can protect te rat's retina which was injured by acute intraocular hypertention.This protective role may be concerned with that it can promote the expression of endogenous BDNF in retina.
