Objective:To investigate the chemotactic activity of CXCL16 on human first-trimester decidual immunocytes(DIC)and its effects on Th1/ Th2 cytokine production of DIC at materno-fetal interface of early pregnancy.Methods:After peripheral blood mononuclear cells(PBMC)and decidual immunocytes (DIC)were isolated and purified,the chemotactic activity of CXCL16 was assessed by Transwell trial.The ELISA kit was used for the determination of IFN-γ,TNF-α,IL-6 and IL-10 in the supernatant of DIC which were cultured for 72 hours with the treatment of rhCXCL16;the quantitative determination of IFN-γmRNA,TNF-αmRNA,IL-6 mRNA and IL-10 mRNA were analyzed by real-time RT-PCR.Results:(1)rhCXCL16 exhibited chemotactic activity on peripheral blood mononuclear cells (PBMC)and decidual immunocytes(DIC).The number of PBMC and DIC recruited to lower wells with a concentration of 10ng/ml of rhCXCL16 was 16256 cells and 12702 cells respectively which was more than the control but not significant difference(P>0.05); rhCXCL16 at a concentration of 100ng/ml recruited more PBMCs and DICs than the control(P<0.01)and the neutralizing antibody to CXCL16 inhibited its chemotactic activity(P>0.05).(2)The human first-trimester decidual immunocytes(DIC)were verified to secrete IFN-γ,TNF-α,IL-6 and IL-10 spontaneously and continuously in vitro..(3)rhCXCL16 of different concentration had different influence on the secretion of cytokines.rhCXCL16 at the moderate dose(50~100ng/ml)inhibited the secretion of IFN-γand TNF-α(P<0.05);the concentration of IFN-γin the dose(50ng/ml)group (6.54±1.56pg/ml)was significantly lower than the control group(19.15±0.34pg/ml)(P<0.01);rhCXCL16 of 50ng/ml inhibited the secretion of IL-6(P<0.01);rhCXCL16 at different doses inhibited the secretion of IL-10 significantly(P<0.01).(4)The result detected by real-time quantitive RT-PCR showed that the expression of IFN-γmRNA,TNF-αmRNA and IL-10 mRNA of DIC with the treatment of rhCXCL16(50ng/ml)was significantly lower than that in control group,and the expression of IL-6 mRNA of DIC with the treatment of rhCXCL16(100ng/ml)was higher than that of control group(P<0.05). Conclusion:CXCL16 recruited and reserved immunocytes into the deciduas and play a role in the secretion of Th1/ Th2 type cytokine at the maternal-fetal interface.
