Analyse The MHC Restricted And Non-MHC Restricted Killing Activity In The Effect Of Cytokine-induced Killer Cells Against Gastric Cancer

Objective:To investigate MHC restricted and non-MHC restricted killing activity in the role of cytokine induced killer cells(CIK) against gastric cancer(SGC-7901) in vitro.Methods:Peripheral blood mononuclear cells(PBMC) from healthy donors were induced to become dendric cells(DC) and CIK,use tumor antigen extracted from gastric cancer SGC-7901 cells to sensitize DC,DC induces cik to become CIK induced by DC(DC-CIK) and CIK induced by gastric cancer antigen-pulsed DC (antigen-DC-CIK).On the 14th day of culture,CIK,DC-CIK and antigen-DC-CIK cell phenotypes CD3,CD3/CD8 and CD3/CD56 ware analyzed by flow cytometry.Take CIK, DC-CIK and antigen-DC-CIK as the effector cells,take the SGC-7901 cells which have or not have the MHC molecule blocked as the target cells.The cytotoxic activity was detected by MTT.Results:On the 14th day of culture,the proportion of CD3~+ CD8~+ and CD3~+ CD56~+ double positive cells in antigen-DC-CIK ware significantly higher than that in DC-CIK and pure CKI(P<0.01).At 20:1 effector-target ratio in vitro,the cytotoxic ability of CIK group,DC-CIK group and antigen-DC-CIK group against SGC-7901 was (46.6±1.3)%,(52.6±0.9)%and(72.5±1.2)%(P<0.01);After SGC-7901's MHC molecule was blocked,the cytotoxic ability was(44.3±0.6)%,(49.9±0.5)%and (50.4±1.1)%respectively.The proportion of MHC restricted killing effect in antigen-DC-CIK was higher than that of DC-CIK and CIK.Conclusion:The major cytotoxic effect of CIK against gastric cancer is non-specific killing effect,MHC restricted killing effect only accounts for 4.9%;When induced by DC, the total cytotoxic effect was enhanced and the major cytotoxic effect is non-specific killing effect,MHC restricted killing effect was enhanced slightly accounts for 5.1%; After induced by antigen-pulsed DC,the major cytotoxic effect is still non-specific killing effect and the specific killing effect was largely increased to 30.5%.
