PART1 ESTABLISHMENT OF RAT AORTIC CALCIFICATION AND ITS INHIBITORY MODELObjective:To establish the model of rat aortic calcification and test its inhibitory model by using Doxycycline.Methods:Rat aortic calcification model was established by subcutaneous injection of Cholecalciferol. And then the inhibitory model was established by both Cholecalciferol and Doxycycline.Results:We can see the morphology in aortic calcification tissues was seriously damaged by both HE and Von Kossa staining. Compared to the experimental group,the degree of damagement of inhibitory group was decreased.Conclusions:The correct methods of configuration and injection is the key to successful establishment of rat aortic calcification and its inhibitory model. PART2THE EFFECT AND CLINICOPATHO-LOGIC SIGNIFICANCE OF TGF-β1 AND DOXYCYCLINE IN AORTIC CALCIFICATIONObjective: To study the expression of transforming growth factor-beta 1(TGF-β1)in different stage on experimental and inhibitory rat aortic calcification and to discuss the possible effects and correlations among TGF-β1,doxycycline and the development of arotic calcification.Methods: Aortic calcification model rats were setup by cholecalciferol. And then aortic calcification inhibition model rats were setup by both cholecalciferol and doxycycline. In all the tested animals,their aortic tissues were obtained for pathological observation and immunohistochemistry staining in different stage. The expression degree of TGF-β1 and the changes of morphology in aortic tissues were measured by image analysis system.Results: Experimental and inhibitory rat aortic calcification models were successful constructed. The expression of TGF-β1 was gradually increased in the different processes of experimental and inhibitory Aortic calcification rats,respectively(P<0.05). Compared to the normal control group,The expression of TGF-β1 of experimental group was significantly increased(P<0.05).But the expression of inhibitory group was significantly lower than that in the the experimental group in the same stage.Conclusions: TGF-β1 may play an important role in the development of aortic calcification. It can be regarded as one of the important markers to predict the severe degree and consequerce of aortic calcification. Doxycycline can inhibit the processes of rat aortic calcification and decrease the expression of TGF-β1 .Our results provide theory foundation for us to study aortic calcification and the mechanism of inhibiting it.
