| Background and Objective Graves disease (GD)is more common of one of autoimmune thyroid diseases in our country. The exact pathogenesis is still unknown so far, but it's generally considered that the interaction of a large number of genetic factors and environmental factors led to the occurrence of Graves'disease. Environmental factors in which iodine intake and genetic factors in HLA-II gene polymorphism and the occurrence of GD were closely related to. At the same time, certain types of HLA-II alleles for GD have a certain genetic predisposition and resistance, and the existence of ethnic and regional differences. Therefore, by studying this subject in patients with GD in coastal areas of Guangxi iodine intake and HLA- DQA1,DQB1,DRB1*09 gene polymorphism and explore the coastal areas of Guangxi GD susceptibility gene and susceptibility to environmental factor.Methods Using sequence-specific primer - polymerase chain reaction (PCR-SSP) detected 162 cases of patients with GD in coastal areas of Guangxi and 90 normal cases of the HLA-DQA1,DQB1,DRB1* 09 genotype; Ammonium persulfate digestion Cerium arsenic (As3 +, Ce4 +) catalytic spectrophotometric determination of all the studies of urinary iodine concentration of contrast. The comparisons of allele frequencies and iodine in urine were made among GD patients and control group.Results (1)The gene frequencies of HLA- DQA1*0501(32.7%),DQB1*0303(35.8%)and DRB1*0901(29.6%) in group of GD patients were significantly higher than the normal control group (10.0%, 12.2 %, 12.2%), the difference was statistically significant (P <0.05); (2) As compared with normal female,the gene frequencies of HLA-DQA1*0301 (44.1%) allele were significantly increased in female GD patients than that in the normal female subjects(26.5%), the difference was remarkable (P <0.05); (3) The median urinary iodine of GD patient group (448.7μg / L) than the control group (219.6.7μg / L) significantly increased. And urinary iodine concentration is higher than 300μg / L frequency distribution of the GD patients were significantly higher than control group, there was a significant difference (P < 0.05);(4) The application of Multi-Logistic regression analysis screening took place with the GD-related risk factors, results showed: gender, iodine intake, HLA-DQA1 * 0501, HLA-DQB1 * 0303, HLA-DRB1 * 0901 and DQA1 * 0301 may be GD risk factors in Guangxi coastal areas (OR, respectively 0.851, 0.971, 0.782 , 0.842, 0.819,0.794).Conclusion (1)For the coastal areas of Guangxi, HLA- DQA1*0501,HLA-DQB1*0303,HLA-DRB1*0901 may be associated with the susceptibility of GD; (2) the HLA -DQA1*0301 allele may be confers susceptibility to female patients with GD in this areas; (3) excessive intake of iodine may be another GD in the region susceptible to environmental factors; (4) It was confirmed that gender, iodine intake, HLA-DQA1* 0501, HLA-DQB1 * 0303, HLA-DRB1* 0901 and HLA-DQA1* 0301were risk factors of GD in Guangxi coastal areas. |
