Objective: laryngeal squamous cell Carcinoma (LSCC)is one of the common tumor in the head and neck. The incidence is rising year after year, The reaserch about it is also increasingly,But the results are not satisfactory. At present Surgical treatment of laryngeal squamous cell carcinoma is one of the important means. The laryngeal squamous cell carcinoma that of invasion and metastasis had a poor effect. Five-year survial rate is lower. At present, There are many methods to evaluate the invasion and metastasis. For example Physical examination, radiographic examination and conventional pathological examination. But they can not satisfy the Clinical diagnosis and treatment for judging the extent of local invasion, neck lymph node and distant organ metastasis. So How to determine a more accurate judgement to invasion and metastasis of laryngeal squamous cell carcinoma, appropriate treatment to improve survival in patients and the quality of life, is an important issue. Study shows that the Mechanism of invasion and metastasis of cancer cells, including tumor cells break through the extracellular matrix (ECM) and vascular basement membrane as well as the formation of tumor angiogenesis and other steps. Matrix metalloproteinases (MMPs) and Tissue inhibitor of mettalloproteinases (TIMPs) are important elements to degradate and remode orderly the extracellular matrix and basement in pathophysiological process. Extracellular matrix and basement membrane degradation are important reasons to invasion and metastasis of laryngeal squamous cell carcinoma and other tumor. This experiment detect the protein and mRNA expression of matrix metalloproteinase 1 (MMP1), inhibitors of matrix metalloproteinase 1 (TIMP1) and inhibitors of matrix metalloproteinase 3 (TIMP3) by immunohistochemistry SP and RT-PCR method in laryngeal squamous cell carcinoma and adjacent tissues. It investigate the relationship between the protein and mRNA expression of MMP1 TIMP1 and TIMP3 and the Clinicopathological parameters. as well as the relationship between eath other. It provide a theoretical basis of defining laryngeal squamous cell carcinoma invasion and metastasis mechanisms and opening up new treatment methods.Methods: The experiment detect the protein expression of MMP1 TIMP1 and TIMP3 by immunohistochemistry SP in 51 cases of carcinoma and 22 cases of adjacent tissues. and mRNA expression of MMP1 TIMP1 and TIMP3 by RT-PCR in 49 cases of carcinoma and 15 cases of adjacent tissues. It analysis the relationship between laryngeal squamous cell carcinoma and clinical pathology parameters as well as the correlation between.ResultS: 1 protein expression:①MMP1 positive expression rates were 82.4% (42/51) and 18.2% (4/22) in laryngeal squamous cell carcinoma and adjacent tissues. Comparison between the two had statistically significant (χ~2=27.156, P<0.05).②TIMP1 positive expression rates were 72.5% (14/37) and 95.5% (1/21) in laryngeal squamous cell carcinoma and adjacent tissues. Comparison had statistically significant (P<0.05).③TIMP3 positive expression rates were 80.4%(41/51)and 100%(22/22) in laryngeal squamous cell carcinoma and adjacent tissues. Comparison had statistically significant (P<0.05).④MMP1/TIMP1/TIMP3 positive expression and histopathological grading, lymph node metastasis was significantly (P<0.05), age had not statistically significant (P>0.05).⑤MMP1/TIMP1/TIMP3 positive expression in laryngeal squamous cell carcinoma had not correlation and statistically significa (P>0.05). 2 gene expression:①MMP1mRNA expression quantity in laryngeal squamous cell carcinoma (0.66±0.26) was higher than adjacent tissues (0.49±0.23) (P<0.05);②TIMP1mRNA expression quantity in laryngeal squamous cell carcinoma (0.55±0.33) was lower than that in adjacent tissues (0.76±0.35) (P<0.05);③TIMP3mRNA expression quantity in laryngeal squamous cell carcinoma (0.34±0.21)was lower than that in adjacent tissues (0.49±0.24) (P<0.05);④MMP1 / TIMP1 / TIMP3 mRNA expression and histopathological grading, lymph node metastasis was significantly (P<0.05), age had not statistically significant (P>0.05).⑤MMP1/ TIMP1/ TIMP3 mRNA expression in laryngeal squamous cell carcinoma had not correlation and statistically significant (P>0.05).Conclutions: 1 MMP1 expression in laryngeal squamous cell carcinoma was higher, But the TIMP1 and TIMP3 expression was lower . It indicated that MMP1 TIMP1 and TIMP3 was involved in tumorigenesis.2 The expression of MMP1/ TIMP1/ TIMP3 were correlated with pathology classification, lymphatic metastasis, indicating that it might play an important role in invasion and metastasis of laryngeal squamous cell carcinoma.
