| Folium Isatidis is the the dried leaf of Isatis indigotica Fort.which commonly used for Qingrejiedu of traditional Chinese medicine in clinical application for many years,with significant effect. Modern study also proved that Folium Isatidis has a various effects made by a variety of chemical components,multi-linked and multi-targeted in the body. But so far,study of Folium Isatidis on whether chemical or pharmacodynamics is respectably scattered and lack of systematic,its effective substance is still not clear and hard to thoroughly clarify its mechanism. These make either the quality control of the materia medica and the compound preparation has no scientific and objective methods and targets. So it is important to find out the active ingredient related to the Qingrejiedu effect, to evaluate and control the quality of Folium Isatidis and its compound preparation.Objective: To find out the effective control indicators which can reflect the inherent quality of Folium Isatidis and its preparation on the guide of effectiveness in real terms of Qingrejiedu, and to establish the new chemical evaluation method of the quality of Folium Isatidis.Methods: Compared the difference between the blank serum and serum containing drug, the experimental first drawn a pointer chromatography of the components in the serum after administration of Folium Isatidis on the basis of means and methods of medicine chemistry,using the HPLC-DAD technology. And studied the correlation between the pointer chromatography and effect on the basis of the build classical model (rabbit fever caused by endotoxin).Then used macroporous resin,silica and HPLC / MS,NMR to isolate and identify the active ingredients in Folium Isatidis. This article had a comprehensive assessment of the quality of Samples from 6 diferent areas on character,identification,inspection and contents determination of anthranilic acid,syringic acid,indigo and indirubin, the 4 indicators come from 2 main categories in Folium Isatidis.Results: The study found that a main component in the blood had a good spectrum–effct correlation with the inhibition of the rabbit fever caused by endotoxic. Compared the spectrum of UV and HPLC chromatography, in the same retention time; there was a higher content of the characteristic peak both in the chromatography of serum containing drug and the drug decoction. And their UV scanning had the same pattern; the maximum absorption is around 250 nm. It is more likely that the two substances are the same substance.This experiment isolated that component based on its physical and chemical propertiesand, named "DQY251", and identified it as tryptamine. Through comprehensive assessment of the quality of Samples from 6 diferent areas we established the new chemical evaluation method of the quality of Folium Isatidis on anthranilic acid,syringic acid,indigo and indirubin.Conclusion: The experimental studied on the anti-endotoxin active ingredient in Folium Isatidis from the serum with the method of Serum Pharmacochemistry and got a main component– tryptamine. The method is scientific,reasonable,rigorous and innovative designed. And we established the new chemical evaluation method of the quality of Folium Isatidis; found that the Folium Isatidis from Bozhou Anhui has the best content and uniformity in the Folium Isatidis Samples from 6 diferent areas, made a more scientific and rational quality evaluation of Folium Isatidis. |
