OBJECTIVE: To investigate the mechanism of pharmacology of Smilax china L.,optimize the extraction method and solution for the extraction of Smilax china L. and study the constituents of Smilax china L..METHODS: Using the coliform bacterium to manufacture rat's chronic pelvic inflammatory disease (CPID) model.Determine the change of IL-2 level in blood serum by using enzyme linked immunosorbent assay (ELISA),examine the contents of MDA in blood serum by using the method of TBA,observe the changes of rat's womb organizes by using Hematoxylin and Eosin Staining method.Using different extraction methods such as traditional extraction and microwave extraction approach to extract the effective components in Smilax china L.,at the meanwhile under the same extraction method using different solvent,n-butanol,acetoacetate etc. to extract the effective components in Smilax china L..Through observing the change of calcineurin (CaN) vigor,compare different extraction methods' extraction ratio as well as different extraction part's effective component to the influence of CaN. Extract and isolate the effective components in Smilax china L. by using TLC, Silica gel column chromatographic analysis, Gelatin column chromatographic analysis, HPLC etc.RESULTS: The level of IL-2 and MDA in rat's blood serum restore to normal ,at the same time pathological section demonstrates that the rat's chronic pelvic inflammatory disease improved by treating and restore to normal.Microwave-assisted extraction has higher extraction ratio than traditional extraction method. The active components extracted by Microwave-assisted extraction have stronger inhibited activity on CaN than that extracted by traditional extraction method.All extractions extracted by different solution could suppress the activity of CaN, and the sequence is:n-butanol>EtAc>alcohol>water. CONCLUSIONS: Smilax china L. has obvious therapeutic action to CPID and might be better than the control group in effection. The function mechanism possibly relates to adjust the immune factor IL-2 and resists the free radical in vivo. How to Smilax china L. display immunosuppression mechanism possibly relates to participating the suppression of CaN activeness.The active site mainly concentrates in the n-butanol extract spot. Microwave-assisted extraction has many merits,such as reducing extraction time, saving production cost and so on, moreover the effective components extracted by microwave has the stronger inhibitory action to CaN.
