OBJECTIVE Curcumin,the major yellow pigment extracted from turmeric,have been reported to have several pharmacological effects. Resently,its anti-tumor effect seem to raise the most interest. Curcumin demonstrates poor systemic bioavailability after p.o. dosing,which may be related to its hard dissolution,inadequate absorption and avid metabolism. So it is necessary to be exploited and developed the new dosage form to improve its solubility and to raise its bioavailability. This study, we use curcumin solid dispersion to evaluate its inhibitory effects on tumors in vivo.METHODS 1 . Growth inhibition rates of cells were measured with MTT method. 2. Tumor inhibition were detected by tumors transplanted subcutaneously in mice with cur-SD(200,100,50,25mg/kg/d)p.o. dosing. 3. The expressions of CD34 and VEGF were assessed by immunohistochemical study. 4.The expression of VEGF,AKT,pAKT and other signal protein molecules ,such as NF-κB andβ-catenin were examined by Western Blot.RESULTS: 1.The Curcumin solid dispersion showed inhibitory effects on different tumor cell lines in a does dependent manner with IC50 values from 6.6 to 12.12μg/mL. 2.In vivo study, the inhibitory rate of curcumin solid dispersion 200mg/kg/d group on H22 and B16 were 43.2% and 53.1%, and 100mg/kg/d group on SW480 was 57.7% ,higher than the same dose of cur group.3. Compare with the control group,the expression of CD34 and VEGF were decreased in high dose group (P<0.01) and the level of AKT,pAKT,β-catenin and other signal protein,such as NF-κB were down-regulated.CONCLUSION:The anti-tumor effects of Curcumin solid dispersion was higher than Cur in vivo,and the most significant effect was on SW480 transplanted subcutaneously in nude mice. Significant anti-tumor effect can be achieved by inhibiting the expression / activity of VEGF or by blocking the down stream signal PI3K/Akt pathways andβ-catenin.
