Objective To investigate the impact of different time bone marrow mesenchymal stem cells transplantation on the occurrence and development of diethylnitrosamine-induced SD rat liver cancerMethods MSCs of male SD rat were isolated and purified in adherent culture in vitro.The forth passage cells (5×106),expanded by subculture,were respectively injected into the homogeneous female rats via the caudalis vena in the ninth week and the thirteenth week of the liver cancer model induced by diethylnitrosamine in rats in the intervention groups.The medium(L-DMEM) was respectively injected into the homogeneous female rats via the caudalis vena in the ninth week and the thirteenth week of the liver cancer model induced by diethylnitrosamine in rats in the control groups.In the eighteenth week of the model,all the living state of rats were killed. The mortality and the incidence of liver cancer in the intervention groups and control groups were evaluated.The pathology of the liver in the intervention and control groups were evaluated by Scheuer inflammation activity score and fibrosis grading score respectively.The degree of liver cell differentiation in the intervention and control groups were also evaluated by Edmondson score.The expression ofα-smooth muscle actin protein and cell cycle of the liver of rats in each group were detected.Result With MSCs transplantation in 9th and 13thweek of the model, no difference in.the mortality,the incidence of liver cancer and the degree of liver cell differentiation was found between the intervention groups and control groups(P>0.05).With MSCs transplantation in 9th week of the model ,there was a statistical difference in the activity of liver inflammation,the degree of liver fibrosis and the expression ofα-smooth muscle actin protein between the intervention groups and control groups(P<0.05).With MSCs transplantation in 13th week of the model,there was no statistical difference in the activity of liver inflammation,the degree of liver fibrosis and the expression ofα-smooth muscle actin protein between the intervention groups and control groups(P>0.05).With MSCs transplantation in 9th and 13th week of the model,there was a significantly statistical difference in the proportion of G1 and S phase cells between the intervention groups and control groups(P<0.01).Conclusion 1)MSCs of young male SD rat were successfully isolated and purified in adherent culture in vitro.It is a way to obtain enough homogeneous MSCs colony.2)In the short term,MSCs transplantation does not help to reduce the mortality,the incidence of liver cancer and the degree of liver cell differentiation. But it can inhibit the proliferation of liver cancer.Early MSCs transplantation contributes to reduce the activity of liver inflammation,the degree of liver fibrosis and the activity of hepatic stellate cells.
