PurposeTo investigate the synergic effect of BMP-14 and bFGF in inducing chondrogenesis of rat bone marrow stem cells.MethodWe use density gradient centrifugation plus adhesion separation to isolate and purify rat BMSCs.BMSCs were cultured in L-DMEM containing 10%fetal bovine serum until the third generation as seed cells.Four study groups were set.Group A is negative control group.100ng/mlBMP-1,10ng/ml bFGF and(100ng/mlBMP-14+10 ng/mlbFGF) were seperately added to group B,C and D.After 24 hours of culture,a comparison was made between four groups for cell proliferation.After 14 days of cultures,observations were made for morphological changes,type-Ⅱcollagen and Alcian blue staining.ResultAfter 24 hours of cell culture,the absorbance(A) values were 0.4430±0.0297 in group A,0.5480±0.0370 in group B,0.6418±0.0534 in group C,0.6764±0.0417 in group D,in addition to C,D no statistical significance between groups(P>0.05),the remaining differences between the groups was statistically significant(P<0.05);After 14 days of cell culture,the positive rate of type-Ⅱcollagen staining was 7.5210±3.7243%in group A,40.8271±1.2787%in group B,17.2818±1.4242%in group C,60.5114±1.7634%in group D.There is statistically significant difference in each group(P<0.05);As for Alcian Blue staining,group A was negative,group B and C were slight-stained,and group D was blue.ConclusionThe combination of BMP-14 and bFGF induces proliferation and chondrogenesis more effectively than either BMP-14 or bFGF alone.
